Objective To observe treatment of Tanshinone IIA, and mixture of Tanshinone IIA and salvianolic acid B indifferent proportionson tumor necrosis factor TNF-a and nuclear transcription factor NF-κ B p65of the ischemic brain tissue of artery occlusion rat model, and to explore their inflammation inhibitory mechanism after cerebral ischemia reperfusion.Methods The rat focal cerebral ischemia-reperfusion injury model was established by suture occlusion of middle cerebral artery. HE staining and immunoh is to chemical methods were applied to observe the pathological changes of ischemic brain tissues, TNF-α and NF-κ B p65.Result HE staining cell necrosis model group is the most obvious and visible nuclear condensation and fragmentation are present. Tissue necrosis of the medication treatment group is less serious compared with the model group. For the sham group and blank group, the nuclei is complete with clear nucleolus, cytoplasm is lightly stained and the boundary of cells and mesenchyme. To compare with the tanshinone IIA group, the group treated by mixture of Tanshinone IIA and salvianolic acid B indifferent proportions has lighter necrosis, with the most obvious one, the tanshinone IIA30mg/salvianolic acid B300mg group. Compared with the normal groupand sham group24h after rat cerebral ischemia, expression of the cerebral TNF-α and NF-κ Bp65of the model group is significantly enhanced even in the48h. Compared with the model group, the TNF-α and NF-κ Bp65around the cerebral lesion of the medication group is significantly reduced. Compared with the same period of the tanshinone IIA group, the reduction is more obvious, with the tanshinone ⅡA30mg/salvianolic acid B300mg group as the most obvious one.Conclusion TanshinonellA and mixture of different proportions of Tanshinone ⅡA/salvianolicacid B can inhibit the inflammatory factor TNF-a and the transcription factor NF-κ Bp65expression, thereby reducing the inflammatory damage after ischemic injury. The effect of a mixture of different proportions of Tanshinone IIA/salvianolic acid B is more obvious, and the tanshinone ⅡA30mg/salvianolie acid B300mg group is the most obvious.
