Objective To observe the influence of Tanshinone IIA and its different proportioning with Salvianolic Acid B on the expression of the inflammatory factors ICAM-1and MMP-9in the ischemic cerebral tissue of the rat middle cerebral artery occlusion (MCAO) model and to discuss its machanism of inhibiting the inflammatory response after the cerebral ischemia reperfusion。Method The cerebral artery was blocked with intraluminal thread technique and the models of rat focal ischemia reperfusion were established in rats. The Hematoxylin-eosin (HE) staining and Immunohistochemistry (IHC) were implemented in order to observe the pathological change in the ischemic cerebral tissue and the expression of inflammatory factors ICAM-1and MMP-9。Result In the model rat groups, the necrosis of neurocytes in the cortical areas was the most significant and the karyopyknosis and.karyorrhexis were discovered as well. In the TSNIIA group, TSMIIA30/SAB100group and the TSNIIA30/SAB300group, the necrosis of neurocytes in the cortical areas was comparatively not as severe as that of the model group. In the sham operated group and blank group, the karyon remained complete and the nucleoli was clear and the nuclei were pale. The boundary of cells and mesenchyme was clear.24hours after the ischemic injury in rats, in the model group, the cerebral expression of ICAM-1and MMP-9increased remarkably and was still increasing after48hours. Compared with the model group, in the TSNIIA group, TSNIIA30/SAB100group and the TSNIIA30/SAB300group, the expression of ICAM-1and MMP-9in the periphery of the cerebral injured area was reduced. In the TSNIIA group, TSNIIA30/SAB100group and the TSNIIA30/SAB300group, the necrosis of tissue was less severe, especially the TSNIIA30/SAB300group。Conclusion Tanshinone IIA and its different proportioning with Salvianolic Acid B is able to reduce the inflammatory damage after the ischemic injury by inhibiting the expression of inflammatory factors ICAM-1and MMP-9. The effect of Tanshinone IIA with its different proportioning with Salvianolic Acid B is even better than Tanshinone IIA, especially the TSNIIA30/SAB300。...
