Objective:Observed the protective effect of peroxisome proliferator activated receptorγ agonist pioglitazone on focal cerebral ischemia reperfusion injury, study the influence of pioglitazone on serum interleukin-6level and the endogenous janus kinase2/signal transducer and activators of transcription3signal transduction pathway, explore its brain protective mechanism.Methods:Using the modified suture method to make focal brain artery ischemia and reperfusion rats model. Experimental animals were divided into sham group, ischemia-reperfusion injury model group, saline intervention group, pioglitazone intervention group. Nerve function scores were used to observe the impact of the PPARγ agonist pioglitazone on rat behavior score, HE staining to observe the changes in rat brain tissue injury, ELISA to observe the impact of PPARγ agonists on serum Il-6content, Immunohistochemical and western-blotting was observed the influence of PPARγ agonists on P-JAK2, P-STAT3expression in cerebral ischemic and reperfusion district. Analysis the relationship between serum Il-6levels and P-JAK2, P-STAT3expression amount in cerebral ischemic and reperfusion district.Results:PPARγ agonist pioglitazone can reduce behavior scores of cerebral ischemia and reperfusion injury rats (P<0.05), reduce brain cell necrosis, and lower serum IL-6levels (P<0.05),decreases P-JAK2, P-STAT3expression in the cerebral ischemia and reperfusion district (P<0.05). Serum IL-6levels and P-JAK2, P-STAT3expression in ischemic and reperfusion district was a positive correlation, respectively, significant at the0.01level (bilateral).Conclusions:PPARy agonist pioglitazone has a protective effect against focal cerebral ischemia and reperfusion injury;The cerebral protective effects of pioglitazone may be associated with lower IL-6levels in the serum; Cerebral protective effect of pioglitazone may be associated with lower cerebral ischemia and reperfusion district JAK2/STAT3signal transduction protein expression; The cerebral protective effect of pioglitazone may be associated with the downward IL-6-JAK2/STAT3signal transduction pathway.
