Objective:To investigate protective effort of Valsartan pretreament on immature myocardium under cardiopulmonary bypass in dog.Methods:Sisteen healthy ang homogeneous minor dogs were randomly divided into two groups, the observed group offer Valsartan (10mg/kg/d) one week in preoperative, the control group offer physiological saline (10mg/kg/d) one week in preoperative, routine establish open-heart surgery on beating heart under cardiopulmonary bypass animal model. The blood sample were taken in different time (CPB10minutes befer、60min after the start of the aortitis、120min after the start of the aortitis) and the changes of cTn-I、TNF-α were measured and in relevant time collect homo-cardiac-tissue (0.5cm3) fixation in fixation fluid,routine paraffin embedding、slice, SP immunohistochemistry test Bax、Bcl-2protein express.Results:The level of cTn-I、TNF-α in the control group increased gradually during CPB, which were especially higher after the recovery of perfusion. The level of cTn-I、TNF-α in the observed group also increased during CPB, but lower that in the control group and there was no increasing trend after the recovery of perfusion. The observed group Bax protein express lower than the control group, and the Bcl-2protein present opposition result.Conclusions:Valsarton pretreatment through reduce mediators of inflammation release, recede Bax protein express, improve Bcl-2protein express, restrain cardiac muscle apoptosis, availably avoid ischemia reperfusion injury, may can offer preferable myocardial protection process.
