ObjectiveTo detect the expressions of NGX6and vascular endothelial growth factor (VEGF) in gastric carcinoma tissues and normal gastric mucosa by immunohistochemical technique and reverse transcription polymerase chain reaction (RT-PCR) in this study, analyze the relationship between the rezult of expressions and the ability of invasion and metastasis of tumor cells, the relationship between the expressions and the biological behavior of gastric carcinoma. So it provides the theoretical foundation for prognosticating the invasion and metastasis of gastric carcinoma, evaluating the prognosis and guiding the reasonable comprehensive treatment of gastric carcinoma.MethodsIn this study,120samples of gastric carcinoma were selected, who had taken the radical operation and proved by pathology in the First Hospital Affiliated to Xinxiang Medical University from Aug.2010to Jun.2011, include gastric cancer tissue and corresponding normal gastric epithelial tissues from over5cm. None of them underwent preoperative chemotherapy and radiation therapy. The expressions of NGX6and VEGF in gastric carcinoma tissues and normal gastric mucosa were detected by immunohistochemical technique and RT-PCR.The relationship between the expressions of NGX6and VEGF and the biological behavior of gastric carcinoma were analyzed, including sex, age,tumor location, size, gross type, differentiation degrees, depth of invasion, lymph node metastasis and TNM stage. And the correlation between NGX6and VEGF in gastric cancer tissue was used by Spearman methods. Results1. The expression of VEGF protein in gastric carcinoma was70.00%(84/120), in normal gastric38.33%(46/120), and the difference between the2groups was statistically significant(p<0.01).2. The positive expression rate of NGX6in gastric carcinoma was43.33%(52/120),in normal gastric tissue87.50%(105/120), and the difference between the2groups was statistically significant(p<0.01).3. The positive expression of VEGF were related to the different tumor diameters, degree of differentiation in different tissues and different depth of tumor invasion, TNM stage and lymph node metastasis,and the difference was statistically significant(p<0.05), but in sex, age, tumorl ocation, size, gross type between the2groups no significant difference(p>0.05).4. The lower expression of NGX6were related to different tissues and cell differentiation, the different depth of tumor invasion,TNM stages and whether the different lymph node metastasis, and the differences were statistically significant(p<0.05), but in sex, age, tumor location, size, gross type, diameter between the2groups no significant difference (p>0.05).5. NGX6protein and VEGF protein in gastric carcinoma were negatively correlated,the correlation coefficient r=-0.367(p>0.01).6. Normal gastric epithelial tissues VEGF mRNA, RT-PCR products after electrophoresis with dark, lower output; in gastric carcinoma product band intensity, higher output, and the optical density was significantly higher compared with the normal gastric epithelial tissues (p<0.05). And normal gastric epithelial tissues NGX6mRNA, RT-PCR products after electrophoresis with intensity, higher output; in gastric carcinoma product band dark, lower output, and the optical density was significantly lower compared with the normal gastric epithelial tissues (p<0.05).And VEGF mRNA expression in gastric carcinoma tissues compared with normal gastric tissue increased,VEGF mRNA expression in gastric carcinoma was (0.70±0.14), in normal gastric tissues expressed as (0.25±0.13)(p<0.05). And NGX6mRNA expression in gastric carcinoma tissues compared with normal gastric tissue decreased,NGX6mRNA expression in gastric carcinoma was (0.31±0.11), in normal gastric tissues expressed was (0.56±0.09)(p<0.05).Conclusions1. Higher expression of VEGF and lower expression of NGX6existed in gastric cancer, suggesting that NGX6and VEGF are related with the occurrence of gastric carcinoma development and transfer.2. Higher expression of VEGF and lower expression of NGX6are related to the histological differentiation, depth of tumor invasion, TNM stages and lymph node metastasis, suggesting that NGX6and VEGF are useful in prognosticating the invasion of gastric carcinoma, evaluating the prognosis and guiding the reasonable comprehensive treatment.
