Epithelial ovarian cancer(EOC) is one of the common malignant tumor ofdepartment of gynaecology,its survival rate is increased in recent years,the majorityof whom were diagnosed at late stage,the5-year survival overall rate less than40%.The growth and development of tumor were related closely with angiogenesis,They play a key role in tumor growth,metastasis and diffusion.Vascular endothelialgrowth factor(VEGF)not only increases the tumor vascular permeability and enhancesthe capacity of cancer cells infiltration,but it promotes the proliferation of endothelialcells and induces new vascularization,to promotes both tumor growth and metastasis.Metadherin(MTDH) gene(aka astrocyteelevated gene1,AEG-1)produced withhuman embryonal astrocytes infected by HIV-1or tumor necrosis factor-ɑ,whoseexpression product is MTDH. The studies had found that MTDH is both an oncogeneand an factor of angiogenesis,it plays a vital role in development,invasion andmetastasis in a variety of malignant tumor,such as melanoma,pleomorphicglioblastoma,renal cell carcinoma,breast cancer,endometrial neoplasms,colorectalcarcinoma,thymic carcinoma.However,in these studies,there are few studies on it inEOC,so it for further research.The mRNA and protein levels of MTDH and VEGF indifferent ovarian epithelial tissue were detected with reverse transcription-polymerasechain reaction(RT-PCR) and immunohistochemistry.To evaluate these role andclinical value in EOC. ObjectiveTo explore the expressions and effect of MTDH and VEGF on occurrence andprogress of EOC.To study on the correlation between MTDH and VEGF proteinexpressions in EOC.Materials and Methods1. Study Object42cases of primary epithelial ovarian carcinomatissues (malignantgroup),including25cases of Serous cystadenocarcinoma,10cases of mucouscystadenocarcinoma,7cases of endometrial carcinoma,20cases of benign epithelialovarian neoplasms (benign group) and15cases of normal epithelial ovarian tissues(normal group) were collected from operation patients in department of gynecology ofthe third affiliated hospital of zhengzhou university from June2010to March2013.The age of malignant group ranged from20to66years,the mean age was47.525.43years,None of patients had been treated with radiotherapy andchemotherapy before surgery.2. MethodsThe mRNA levels of MTDH and VEGF In the three groups were detected withreverse transcription-polymerase chain reaction(RT-PCR),The protein levels ofMTDH and VEGF In the three groups were detected with immunohistochemistry.3. Statistics approachThe data were analyzed using the SPSS17.0statistical software,Measurementdata were compared with one-factor analysis of variance,x±s on the behalf ofit.enumeration data was performed with the chi-square test.Pearson’s correlation wereused to analyze the data.P<0.05denotes the difference possessing statisticalsignificance.Results1. The relative expression quantity of MTDH mRNA in malignant group was0.672±0.115,It was significantly higher than that in benign group(0.324±0.041) and in normal group(0.317±0.035) the differences having statistical significances(P<0.0167).2. The relative expression quantity of VEGF mRNA in malignant group was0.714±0.129,It was significantly higher than that in benign group(0.251±0.039)and in normal group(0.243±0.031).the differences was statistical significances(P<0.0167).3. The positive expression rate of MTDH protein in malignant group was69.0%,significantly higher than that in benign group(20.0%) and normal group(13.3%).Thedifference was statistically significant(P<0.0167).4. The positive expression rate of VEGF protein in malignant group was69.0%,significantly higher than that in benign group(20.0%) and normal group(13.3%).Thedifference was statistically significant(P<0.0167).5. The expression level of MTDH gene in epithelial ovarian cancer wassignificantly positive correlated with VEGF gene.6. The expression of MTDH and VEGF in EOC were related to clinical stagesand lymph node metastasis(P<0.05), but not related to pathological typedifferentiation(P<0.05).Conclusions1. The positive expression rate of MTDH in malignant group significantlyhigher than that in benign group and normal group.Our data suggest that expression ofMTDH is associated with ovarian cancer development and progression.2. MTDH have high expression in epithelial ovarian cancer as VEGF inepithelial ovarian cancer,The relationship is a significant positive correlation.Theexpression of VEGF may induced by MTDH through a mechanism.3. Overexpression of MTDH gene was regarded as the poorly prognostic marker.MTDH might as an important transcription factor of angiogenesis,The expression ofMTDH and VEGF is positively correlated with the growth and metastasis of EOC, and there might be a synergy between the two.
