ObjectiveThis study is to investigate the effect of four kinds of porcelain crownsubstrate alloy on the secretion of prostaglandin E2and cyclooxygenase-2byhuman gingival fibroblast.Methods1、 Healthy gingival tissue were obtained from clinicaloperations,and primary culture of HGFs was performed using thetissue culture method. By vimentin and cytokeratinimmunofluorescence staining to identify the cell source.2、Put the nickel-chromium alloy, chromium cobalt alloy, puretitanium and gold palladium alloy into DMEM culture medium forpreparing metal leaching liquid. Cultured human gingival fibroblastsby the metal leaching liquid. With untreated DMEM medium asnegative control, by enzyme linked immunosorbent assaydetermination of prostaglandin E2expression levels, usingimmunofluorescence observation of cyclooxygenase-2expression. Results1、Immunofluorescence result revealed that vimentin staining waspositive and cytokeratin staining was negative. Confirmed cell formesodermal fibroblasts.2、 By ELISA test,HGFs secrete prostaglandin E2levels innickel-chromium alloy group, chromium cobalt alloy group andnegative control group had statistically significant difference(P <0.05), in pure titanium group, gold palladium alloy group andnegative control group had no significant difference (P>0.05).Conclusions1、Successfully established the culture model in vitro of human gingivalfibroblast.2、Nickel-chromium alloy and cobalt chromium alloy extract influence humangingival fibroblasts secrete prostaglandin E2and cyclooxygenase-2. Puretitanium and gold palladium alloy extract have no obvious influence on humangingival fibroblasts secrete prostaglandin E2and cyclooxygenase-2...
