ObjectiveTo explore the exffect of cytokine in Choukroun’s platelet-rich fibrin (PRF) clot stimulates in vitro proliferation and osteogenic and adipogenic differentiation of SD rat bone marrow mesenchymal stem cell.MethodsBMSCs from primary cultures were cultivated with or without a PRF clot originating from SD rat, in proliferation or osteogenic and adipogenic differentiation conditions. After7days, MTT tests in proliferation group. At14days,Alkaline phosphatase (ALP) activity quantification,bone gla proteins (BGP), Alizarin Red S staining and mineralisation nodules counts were performed in osteogenic induction group,and the cells were stained by oil red O in adipogenic induction group.ResultsMTT result show that:from1st day to3rd day,there was no significant difference between PRF group and control group.From5th to7th day,there was significant difference between the PRF group and control group.ALP, BGP,Red S staining and mineralisation nodules counts all show that PRF group was better than control group.Oil red O also show that the formation of lipid droplets in PRF group was more than control group.ConclusionsPRF clot generated a stimulation of the rat BMSC proliferation and both of osteogenic and adipogenic differentiation. It can be the sourse of growth factor for bone tissue engineering.
