ObjectiveTo investigate the human umbilical cord mesenchymal stem cells(HU-MSCs) for treatment of liver cirrhosis in rats, analysis treatment effect indifferent points after the HU-MSCs transplantation, and to investigate themechanisms of the HU-MSCs repair liver cirrhosis.Methods90male SD rats (223.6±20.3g) were selected from liaoning medicaluniversity animal center.24rats were randomly selected as the normal group;The rest66were injected40%CCl4subcutaneous to prepare cirrhotic rat model;after8weeks,10rats were randomly killed, to confirm whether cirhotic modelsuccessful; the remained model group rats were randomly divided into modelgroup and the HU-MSCs transplantation group. After transplantation at the1st,3rd,5th weekend, test the serum for liver function; histopathologicalchanges;immunohistochemistry of hepatocyte growth factor(HGF), transforminggrowth factor-β1(TGF-β1) and anti-human EGF-R antibody expression;enzyme-linked immunosorbent assay test for detection of the serum TGF-β1and HGF.ResultsAfter cirrhotic rat model was successfuled, liver function appear varyingdegrees of abnormal in serum, and liver tissue fake leaflets formation; AtHU-MSCs transplantation the1st,3rd,5th weekend, the HU-MSCs transplanta- tion group AST reduced from202.87±8.54U/L to108.50±3.55U/L, ALT levelsreduced from114.87±6.88U/L to73.75±5.99U/L, ALB increased from19.86±1.69g/L to28.04±1.90g/L, except the first weekend after transplantation,compared with the same period in the model group, the biochemical indicatorsimproved; serum HGF increased gradually after transplantation,which increasedfrom104.51±6.71ng/L to140.94±8.35ng/L, while the model group ratscompared with normal group increased, but no significant difference with thetransplantation group; serum TGF-β1in the HU-MSCs transplantation groupdecreased from117.69±5.42ng/L to65.60±3.48ng/L, while the model serumTGF-β1are still at a high level; after HU-MSCs transplantation, in liver tissueobserved anti-human EGF-R expressi-on, while in the normal group and modelgroup, there were not seen the positive expression of the anti-human EGF-R;fibrosis percentage of HU-MSCs transplantation group decreased significantlyfrom8.14±0.67reduced to2.62±0.36, while the model group significantlyhigher than the normal group, each time there are no significant change.ConclusionsAfter HU-MSCs transplantation, which can improve the impaired liverfunction, and biochemical indicators improved; and observed that the HU-MSCshoming to the lesions of the liver tissue.Through inhibited the expression ofTGF-β1and promoting HGF expression, stimulate liver cell proliferation,repaired liver cirrhosis tissue; so that promote the damaged rat liver return tonormal.
