ObjectiveStudy on the protective effect of SWYSD on degeneration cartilage ofOsteoarthritis(OA) by observing matrix metalloproteinase-1(MMP-1), type Ⅱcollagen in articular cartilage and interleukin-1β (IL-1β) in synovial fluid inanimal model.Methods48male New Zealand white rabbits were randomly divided into blankcontrol group (A group), model group (B group), Diacerein capsule group (Cgroup), SWYSD (D group) after a week of adaptive feeding. Each group were12rabbits. OA animal models(B,C,D group) were made by knee injection ofpapain with reference to Muehleman experiments. Rabbits in C, D group wereinterfered with medicine in2weeks after the last injection of papain. A,B groupwas given normal saline.Samples were taken at5,10weeks after treatment.Synovial fluid was extracted to measure IL-1β content by ELISA in eachgroup.Some cartilage samples were cut to measure MMP-1and type Ⅱcollagen expression level in each group. Some cartilage samples were cut tomake HE staining and Mankin scores of articular cartilage to compare damagein each group.Results1、Comparison of Mankin scoreAt5weeks, the Pathological score in SWYSD group increased and thereis no significant difference, compared with model group(P>0.05). ThePathological score in Diacerein group increased but less than model group(P<0.05). At10weeks,the Pathological score in SWYSD group increased butless than model group(P<0.05), there is no significant difference, compared with Diacerein group.(P>0.05).2、Comparison the expression level of MMP-1and typ Ⅱ collagenMMP-1immunohistochemical staining: At5weeks, the MMP-1inSWYSD group increased and there is no significant difference, compared withmodel group (P>0.05). The MMP-1in Diacerein group increased but less thanmodel group(P<0.05). At10weeks, The MMP-1in SWYSD group increasedbut less than model group(P<0.05), there is no significant difference,compared with Diacerein group (P>0.05).Typ Ⅱ collagen immunohistochemical staining: At5weeks, the typ Ⅱcollagen in SWYSD group decreased and there is no significant difference,compared with model group.The typ Ⅱ collagen in Diacerein group decreasedbut more than model group(P<0.05). At10weeks, the typ Ⅱ collagen inSWYSD group decreased but more than model group, there is no significantdifference, compared with Diacerein grou and there is no significant difference,compared with model group (P>0.05).The IL-1β in Diacerein group increasedbut less than model group (P<0.05). At10weeks, the IL-1βin SWYSD groupincreased but less than model group(P<0.05), there is no significant difference,compared with Diacerein group (P>0.05).ConclusionsThe mechanism of treating OA by SWYSD is to reduce the level ofMMP-1in cartilage to slow down the degradation rate of type Ⅱ collagenandlower the level of IL-1β in synovial fluid to reduce inflammatory response ofarthritis. The effect of SWYSD on OA is significant in long-term.
