ObjectiveTo establish a simple rapid and accurate kinetic monitoring assay fordetecting Glyceraldehyde-3-phosphate dehydrogenase(GAPD) that can beapplied to biochemical automatic analyzer, and evaluate the reliability of themethod of assessment, laid the foundation for future clinical practice.Determination of blood GAPD activity to explore the significance of GAPDin diabetic nephropathy.MethodsUsing triethanolamine buffer (PH7.4),1,3-diphosphoglycerate assubstrate, coupling of3-phosphoglycerate kinase, GAPD reversescatalytic1,3-diphosphoglycerate to3-glyceraldehyde phosphate meanwhilecoenzyme I (NADH) is oxidized to oxidized coenzyme I (NAD+). At340nm ofwavelength NADH has strong absorption peak within the linear range. Theabsorbance of the decline is directly proportional to activity of GAPD.Determining the decline in value of the of the wavelength of340nmabsorbance can calculate the activity of GAPD indirectly. Determination30healthy and80patients of type2diabetes patients,the activity ofGAPD,microalbuminuria (mALB)and β2-micro-globulin (β2-M). Usingstatistical methods analyze the results. ResultsThrough testing several parameters of GAPD to establish a method thatcan be applied to automatic biochemical analyzer. Activity of GAPD inthree disease group were higher than the normal group (P <0.05). mALBandβ2-M in two disease group in the abnormal microalbuminuria group higherthan the control group (P <0.05), but mALB and β2-M in normal albuminuriagroup compared with the control group has no significant difference (P>0.05).GAPD in disease group has no difference (P>0.05), mALB and β2-Mmeaningful differences between the three groups (P <0.05).ConclusionsContinuous monitoring method to determine GAPD is simple and easy tooperate. The kit is very stable, and has high precision and accuracy, widelinear range, and can be applied to automatic biochemical analyzer, easy tospread in the sample measured. GAPD activity in normal proteinuria changedearlier than mALB and β2-M.It can be forecast early diabetic kidney disease,easy to make a diagnosis for clinic, improving the prognosis of patients.
