| Objective:To investigate the effect of vasopressin on p38ã€JNK in MAPK and casepae-3as well asNF-kB during cardiopulmonary cerebral resuscitation in rats.Compared the curative effects ofvasopressinã€epinephrine,and the combination of the two drugs on cardiopulmonary cerebralresuscitation in rats.Methods:Two hundreds and fifty rats(male or female) were divided into five groups randomly(n=50animals each):1ã€Sham-operated group:no asphyxia and no cardiopulmary resuscitation,Only for endotracheal intubation.2ã€Control group: After asphyxia leading to cardiac arrest,start cardiopulmonary resuscitation (CPR) immediately and injected rats1ml of saline.3ã€Epinephrine group: After asphyxia leading to cardiac arrest, start cardiopulmonaryresuscitation (CPR) immediately, and injected rats1ml of epinephrine(0.2mg/kg).4ã€Vasopressin group:After asphyxia leading to cardiac arrest,start cardiopulmonary resuscitation(CPR) immediately and inject ed rats of1ml vasopressin(0.8U/Kg).5ã€Combination group:After asphyxia leading to cardiac arrest, start cardiopulmonary resuscitation (CPR)immediately,and injected epinephrine(0.2mg/kg)and vasopressin(0.8U/Kg)of1ml everythree minutes.Since the lowest survival rate of the control group,it had been abandoned in thefollowing experiment.The rats in the other groups which survived afer CPR, were killed in1hour,3hour,6hour,12hour respectively and the brains had been taken out.We tested theexpression of the JNK〠P38/MAPK,caspase-3, and NF-kBp65in CPCR rats’brains byimmunohistochemical method.Recarded the ROSC rate and observed the pathological changesof the brains in each group.Results:1ã€The ROSC rate between epinephrine group and combination group has statisticaldifference(p<0.05).The ROSC rate between vasopressin group and combination group has no statistical difference(p>0.05).2ã€Compared to the epinephrine group,the expressio ns of the JNK, P38MAPK, caspase-3and NF-kBp65in the vasopressin group and combination group are lower, and the difference has statistics significance (p<0.05).Although the expression of the JNKã€P38ã€caspase-3ã€NF-kBp65are lower in the combinationgroup,the difference between vasopressin group and combination group has no statisticssignificance(p>0.05)3ã€Observed after HE staining, morphology changes are not veryobvious among the three groups of epinephrine group,vasopressin group and combination group under light microscope.Compared to the Sham-operated group,neuronal apoptosis and neurons edema can be seen in the others.But under electron microscopy ultrastructure,we can see the damage of the epinephrine group is more serious than vasopressin and combination group.It is not obvious that the damage between the latter two groups.Conclusion:The ROSC rate between epinephrine group and combination group has statisticaldifference(p<0.05).The ROSC rate between vasopressin group and combination group has nostatistical difference(p>0.05). Through the comparison of immunohistochemical results andpathology result,we can draw the conclusion that the vasopressin can increase the cerebralblood flow perfusion and reduce the apoptosis of neurons.Also can it suspress the MAPK andNF-kBsignaling pathways and alleviate the ischemia reperfusion injury. |
