Fluorescence spectra of α-asarone and β-asarone, active components in traditionalChinese medicine, were studied in this thesis. Influence of experimental conditions onfluorescence spectra was investigated. A high performance liquid chromatographic method byusing FLD detector was proposed for the determination of α-asarone and β-asarone inRhizoma Acori Tatarinowii (RAT, Shichangpu in Chinese) and Fructus Carotae (FC, Nanheshiin Chinese) respectively. In addition, a three-dimensional fluorescence method wasestablished for the identification of RAT and FC from their counterfeit drugs.Three parts were included in this thesis:1. Fluorescence spectra of α-asarone and β-asarone were studied. The influence of ionicstrength, acidity, solvent, surfactant effects on spectral properties was inspected. The linearrelationship between fluorescence intensity and concentration was also investigated. Thefluorescence quantum yields of α-asarone and β-asarone were measured to be0.609and0.646respectively, using quinine sulfate as a reference.2. Fluorescence property of the methanol extracts of RAT and FC were studied. Thecontent of two types of asarone in RAT and FC were separately determined by highperformance liquid chromatographic method with diode-array detector (DAD) andfluorescence detector (FLD). Experiments showed that the results of the two detectors arebasically the same. But, compared to DAD detector, FLD detector has the advantages ofhigher sensitivity and lower detection limit. This work provided a new approach for thedetermination of asarone in traditional Chinese medicine.3. Three-dimensional fluorescence fingerprints of RAT, FC and their counterfeit drugswere tested, thereby a3D fluorescence examination method for the identification of thesemedicinal herbs was established.
