objectiveThrough the observation of the different effect of separate and joint use of nervegrowth factor and nimodipine use of rat sciatic nerve injury of peripheral nerve injuryregeneration modelon sciatic nerve function, nerve electrophysiology andmorphological index of rat sciatic nerve injury regeneration model and comparing thedifferent effect for the two drugs combined treatment of different ratio, dosage time,explore the effect combined treatment on the regeneration of peripheral nerve injury.methods165SD rats were randomly divided into by experimental demand. all the animalswere made of peripheral nerve injury regeneration model by cutting off the left sciaticnerve and epineurium intermittent suture. Giving medicine groups were givenoperation side intramuscular injection drug0.5ml and control group were given equalnacl. rat sciatic nerve function evaluation and nerve physiological indexesdetermination were regularly done and morphological observation were done after12weeks.results1. Nerve growth factor low and high dose (1u/kg;4u/kg) groups are superior tothe control group (P <0.05) in sciatic nerve repair function, nerve electrophysiologyand morphological index, high dose group in nerve electrophysiology index andmorphological index is superior to small dose group(P <0.05).2. Nimodipine low and high dose (125μ g/kg;500μ g/kg) groups are superior tothe control group (P <0.05) in sciatic nerve sciatic nerve repair function, nerveelectrophysiology and morphological index, low group is better than that of thecontrol group (P <0.05) only in nerve electrophysiology index, high dose group issuperior to small dose group (P <0.05) in nerve electrophysiology index andmorphological index.3. Nerve growth factor and nimodipine joint dosage dose groups are superior tothe control group (P <0.05) in sciatic nerve repair function index, nerveelectrophysiology index, the Combined treatment high-dose group is superior to small dose group (P <0.05) in neural electrical physiological index.4. comparing from single low dose nerve growth factor or nimodipine groups theelectrical physiological indexes NCV of the same dose Combined treatment groupsincrease obviously, and the same results are shown in high dose groups.Alone withhigh dose of drug nerve growth factor or nimodipine groups comparison, electricalphysiological indexes NCV and sciatic nerve function index of large dosecombination drug groups has significantly improved (P <0.05).5. To electrical physiological indexes NCV as observation indexes, the singledose and joint dosage groups data are made variance factorial analysis and foundthat small dose of nerve growth factor and large doses of nimodipine joint useproduces obvious interaction.6. To electrical physiological indexes NCV as observation index, postoperativeimmediately giving medicine is superior to the effect of postoperative24hours or48hours.Conclusion1. Joint and separate application nerve growth factor and nimodipine has obviousrole in promoting on rat sciatic nerve injury recovery regeneration model, and presenta certain concentration dependence.2. Combined treatment promoting function of rat sciatic nerve injuryregeneration model is better than that of the single application nerve growth factorand nimodipine in the same dose, small dose of nerve growth factor and large doses ofnimodipine joint use produces obvious synergy.3. Early Combined treatment is much better than late.
