OBJECTIVE:Osteoprotegerin has close relationship with clinical oral orthodontic bone reconstruction, Wnt3a and Wnt5a are related with osteogenesis, but in the orthodontic bone reconstruction has not reported. Through the establishment of the rat orthodontic tooth movement model to observe osteoprotegerin and Wnt3a、Wnt5a expression during experimental tooth movement in rats.METHODS:72male Wistar rats were included, randomly divided into12cage, every cage six rats. According to the volume fraction of chlorine aldehyde hydrated10%of350~400mg/kg anesthetized rats with intraperitoneal injection, will Ni-Ti traction spring at the end of a double wire in the upper left cut teeth, and the other end double wire tied knot with side in rats in the first molar, with50g traction. Orthodontic tooth movement model of rat molar mesial movement was prepared, each health side (not to install orthodontic device side) for blank controls.The model was drawn at1,3,5,7,10and14days tooth movement.36rats used in immunohistochemical study, and others used for RT-PCR.Testing the expression of purpose factors in protein and molecular level. RESULTS:①Strength3d group, the pressure side periodontal lumen narrowed, osteoclast(OC) occurred within near the alveolar periodontal; Pressure side periodontal lumen was narrowed in strength7d group. In the edge of alveolar bone appears obvious bone absorption of silkworm erosion lacuna, there were mononuclear lacuna or more nuclear osteoclast.②The OPG and Wnt3a, Wnt5a are positive expression in immunohistochemist which main role in the cytoplasm shows tan granular shading. OPG positive cells are concentrated in bone formation pressure side position, namely the first molar near periodontal cell and alveolar bone osteogenetic cells. The teeth move1days, rats pressure side OPG expression and there were no differences between the contrast, both have weak positive expression; The teeth move three days, rats pressure side OPG express strengthened; The teeth move5days,7days,10days OPG have strong positive expression in rats pressure side, especially the seventh day; The teeth move14days, OPG express abate. Wnt3a and Wnt5a positive cells express part is not focused, and scattered distribution in periodontal cell and alveolar bone osteogenetic cells.There is no linear relation between time and positive cells expression.③OPG and Wnt3a, Wnt5a in RT-PCR appeared in bands of purpose which is accord with the Immunohistochemical results. Conclusion:In the orthodontic process, OPG involved in the reconstruction process of orthodontic periodontal tissue, OPG in pressure side with teeth mobile expression has increased orthodontic time dependence. It provides a very good point for orthodontic treatment for teeth on the moving velocity and the control of alveolar bone rebuilt aspects. Wnt3a and Wnt5a also have expressed in orthodontic tooth pressure side, but with no obvious relationship between strength time length. The OPG and Wnt3a, Wnt5a expression may be one of the molecular mechanism of orthodontic characteristics. It may be to provide new research direction of orthodontic treatment.
