ObjectiveIn chronic inperiodontitis,detect the expression of Wnt3 a and Wnt5 a,Wnt signaling pathway related ligand.Analysis the correlation between Wnt3 a ligands, Wnt5 a ligand and clinical attachment loss,Wnt5 a ligand and serum TNF-α,to research mechanism of alveolar bone destruction and provide the theoretical basis for regenerative therapy.MethodsResearch object were selected from systemically healthy non-periodontitis(n=30) and chronic periodontitis subjects(n=30). All of Research Object have difference reason,needing to remove the gum tissue.Recording probing depth(PD),clinical attachment loss(CAL),plaque index(PLI) and bleeding on probe(BOP) of all patients and In the operation area, probing depth(PD) clinical attachment loss(CAL) of tooth. Determine the expression of the Fasting plasma glucose(FPG),Glycated hemoglobin(Hb A1c) in plasma samples from all patients.In addition,The m RNA and protein levels of Wnt3 a and Wnt5 a in periodontal tissues were measured by RT-PCR and by Western-Blot,Serum levels of TNF-α were assessed by enzyme-linked immunosorbent assay(ELISA) respectively.SPSS 17.0 package was fitted to this study Results1.There is no difference between periodontally healthy group and chronic periodontitis,revolving age,gender and Hb A1 c,FPG.2.There is statistical significance between periodontally healthy group and Chronic periodontitis, about probing depth( PD),attachment loss( AL),plaque index( PLI) and bleeding on probe(BOP) of all patients( P<0.05).3.The m RNA and protein levels of Wnt3 a and Wnt5 a were significantly increased in the gingival tissues of the chronic periodontitis when compared to the nonperiodontitisgroup(P<0.05).4.There is a negative correlation between the level of Wnt3 a protein and clinical attachment loss(P=0.000,r=-0.764),but positive correlation between the level of Wnt5 a protein(P=0.000,r=0.655).5.There is a positive correlation between the expression of Wnt5 a protein and serum levels of TNF-α(P=0.000,r=0.699).ConclusionsNegative correlation existed between the level of Wnt3 a protein and clinical attachment loss;Positive correlation exists between the level of Wnt3 a protein and clinical attachment loss.Positive correlation exists between the level of Wnt5 a protein and serum levels of TNF-α.Wnt3 a.Wnt signaling pathway promote or inhibit the destruction of the alveolar bone,directly,or by changing the balance of bone metabolism,affecting chronic periodontitis inflammatory process.
