Objective:To study the proliferation inhibition and apoptosis induction effect of ellagic acid on nasopharyngeal carcinoma CNE-2cells. The purpose of this research was to find out the effects and mechanism of EA on nasopharyngeal carcinoma CNE-2cells, and provide possibility to find an effective treatment way to cure NPC.Methods:4groups of nasopharyngeal carcinoma CNE-2cells were cultured in vitro for24h, then intervened by different concentrations (0、2、4、6mg/l) of ellagic acid.1. The morphological changes of CNE-2cells were observed by microscope after intervening by ellagic acid after48h.2. Observe the apoptosis of cells which were stained by Hoechst33258by fluorescence microscope, after intervening by ellagic acid after48h.3. Proliferation of cells was tested by the experimental method of MTT in24h、48h、72h, after intervening by ellagic acid.4. The apoptosis rates of the cells were observed by flow cytometry after48h.5. The expressions of COX-2、NF-κB were tested with Western-blot after the CNE-2cells were intervened by ellagic acid after48h.Results:1. After intervened by different concentrations (0、2、4、6mg/l) of ellagic acid, The higher concentration of ellagic acid, the more phenomena of floating cells, but the fewer living cells left(p<0.05).2. After stained by Hochest33258, The higher concentration of ellagic acid, the more phenomena of pyknosis and karyoclasis, but the fewer CNE-2cells.3. The CNE-2cells was tested by MTT, the higher concentration of ellagic acid, the more clear phenomena of CNE-2cells, growth inhibition.4. After intervened by different concentrations (2、4、6mg/l) of ellagic acid for48h, compared with the control group (0.21%±0.08%), the apoptosis rates of the treatment groups,(2、4、6mg/l)were(19.17%±0.91%、38.44%±3.13%、61.74%±4.28%) different obviously(p<0.05).5. Compared with the control group, the higher concentration of ellagic acid, the expression of COX-2、NF-κB were decreased observly in the treated groups in accordance with concentration.Conclusions:Ellagic acid can inhibit the proliferation of CNE-2cells, induce the apoptosis of CNE-2cells. The potential mechanisms might be to relate to the decreasing expression of COX-2、NF-κB.
