Objective:To explore the inhibitory effects of Ellagic Acid on the proliferation and apoptosis of erythroleukemia K562cells In vitro.Methods:Inhibitory effect on cell growth and induction of apoptosis were examined in vitro.①The erythroleukemia K562cells were treated with EA in0μ g/ml,40μg/ml,80μg/ml,160μg/ml,320μg/ml for24,48,72hours. The ceil viability wasdetected by MTT in all groups.②The cell cycle arrest and apoptosis was detected by flow cytometry when K562cells were treated with EA of diferent concertration inthe48hours.③It Was seen that cells in different degrees of shrinkage, reduction of nuclear fission, and secretory granules in culture medium under the invert microscope④Annexin V-FITC fluorescent staining method the increasing proportion of early apoptotic cellsdisplay.Results:①EA significantly inhibited the proliferation and viability of the K562cells in a time and dose dependent manner compared with the untreated control groups (all P<0.05).②Flow cytometric analysis revealed that treatment with EA resulted in cell-cyclearrest in the G1phase and increasing the percentages of apop totic cells in adose-dependent manner.③typical apoptotic changes was observed by H. E staining, including characteristicchromatin condensation, nuclear shrinkage, nuclear c.lea vage, apoptotic bodies and somecells were floating.④Fluorescence staining showed.160μg/ml ellagic acid arter48hours of cells in the microscope, shrinkage, deformation, chromatin condensation and Marginalized, part of the cell has a smaller volume and a fluorescent fragment or thick with dense granular, massive strong fluorescence. Conclusions:①The growth and proliferation of human prostate cancer K562cell line could be inhibited by Ellagic Acid and its degree of inhibiting depended on its concentration and the time.②Ellagic Acid could induce apoptosis and arrest the G1/S phase in K562cellline.③Ellagic Acid may be used as an anti-tumor drug to induce apoptosis in the chemotherapy of tumor.
