Objective:To establish animal model of guinea pig anaphylactic shock, explore the expression of IL-4, IL-13and IgE in guinea pig serum, provide scientific and objective evidence for clinical and medicolegal diagnosis of anaphylactic shock as well.Methods:30guinea pigs were randomly divided into two groups, the experimental groups with20guinea pigs and the control group with10guinea pigs.0.2ml people pooled serum was injected into the right hind paw of guinea pigs in the experimental group on the seventh day of The experiment to sensitize,3weeks after immunization,1ml people pooled serum was injected into the heart of guinea pigs to induce anaphylactic shock and death. Guinea pigs in the control group were injected with normal saline through the same procedure, and then were put to death by breaking marrow before observing its clinical manifestations.1. Elisa method was used to detect IL-4> IL-13and IgE by extracting serum of guinea pigs in the experiment group and the control group, and SPSS18.0software package was used to analyze the experimental data.2. Chemical staining was applied to observe the degranulation of mast cells.3. HE staining was used to detect the morphological characteristics.Results:1.Symptoms of anaphylactic shock appeared in sensitized group with shock rate of100%and death rate of75%, while there was no expression of anaphylactic shock and death in the control group.2. The guinea pigs’ larynx and trachea were of edema, pulmonary alveolus were of expansion, and alveolar wall became thin with infiltration of eosinophilic granulocyte. There was hydropic degeneration in organs such as liver and kidney.3.Degranulation of mast cells in lung tissues was observed.4. The IL-4levels of serum in experimental group is higher than that in control group, P<0.05; The IL-13levels of serum in experimental group is higher than that in control group, P<0.05; The IgE levels of serum in experimental group is higher than that in control group, P<0.05.Conclusion:1. An animal model of anaphylactic shock was successfully established.2. The level of serum IL-4, IL-13and IgE in the experiment group was obviously higher that in the control group. This provides an evidence for anaphylactic shock forensic identification and clinical diagnosis.
