ObjectiveTo explore the effect of transient expression of human cholesteryl ester hydrolase(hCEH) on CD36and SR-A Ⅰi n TPH-1macrophage.MethodsExperiment was divided into three groups,the control group、ox-LDLinduced group、 hCEH transfection group.TPH-1Cells induced to differentiateinto macrophages.TPH-1macrophage were transfected transiently withpEGFP-N1-hCEH under the stimulation of FuGENE HD.After24h,macrophages were loaded with oxidized low density lipoprotein(100mg/L) for48h. The cellular lipid droplets accumulate was deteced with oil red Ostaining.The expression of pEGFP-N1-hCEH Observed by fluorescencemicroscopy. The CD36and SR-A Ⅰ cells surface protein were determined byWestern Blot. The CD36mRNA and SR-A ⅠmRNA were quantified byRT-PCR.ResultsThere were the largest number of fluorescent cells, after72h transfection,the transfection efficiency of nearly50%. The number of the oil red O stainingtransfection hCEH group compared with ox-LDL group Significantly reduce,CD36, SR-A I receptor protein and mRNA expression was significantly reduced. ConclusionshCEH reduce the expression of CD36and SR-A I in the TPH-1macrophages, inhibition of macrophage foam cell formation.
