ObjectiveTo observe the effect of sodium pyrubate on the kidney vascularpermeability in rats with50%TBSA scalded injury.MethodsMale Sprague-Dawley(SD)(240~260g)rats were divided into four groups:①sham scald,waterbath in37℃.②scald shock with no resuscitation,waterbathin80℃on rat back and both lower extremities15s and abdomen8s,total scaldarea for approximately50%TBSA.③scald shock with NaCl and④scald shockwith NaPyr,intravenous injection of either NaCl or NaPyr with Parklandformula.At4and8h after injury,the rats were killed.HCT and Cr among bloodwere observed,meanwhile kidney vascular permeability was investigated withEvans Blue.Kidney water content was determined with oven drying method.Thecontent of TNF-α were tested with ELISA KIT. The content of MDA were testedwith the TBA spectrocolorimetry.The kidney tissue pathological changes wereobserved.Results1. The rat scald model were established.Scald areas of the rat model were identified as50%TBSA by paper-sticking method. Scald degree was identifiedby Pathology of tissue.2. Effect of NaPyr on kidney vascular permeability of rat scald shockmodel.Compared to that of sham scald group and scald shock with noresuscitation group,water contents of kidney tissues and permeability to EBwere significantly lowered on scald shock with NaPyr group at4h and8hpostinjury(P<0.01).3. Compared to that of sham scald group and scald shock with NaPyrgroup,blood HCT and Cr were significantly higer on scald shock with noresuscitation group and scald shock with NaCl group(P<0.01).4. NaPyr enhances the duration of survival. Both NaPyr and Naclincreased the survival time of scald shock rats, the effects of NaPyr were moresignificantly. The scald shock with NaPyr group had an average time to death of719.8±80.9min, the shock with NaCl group had553.6±68.7min, whereas scaldshock with no resuscitation survived for an average of363.9±47.6min (P<0.01).5. The expression of TNF-α and MDA in scald shock with no resuscitationgroup and shock with NaCl group was obviously higher than those in shamgroup(P<0.01). Compared to that of shock with NaCl group, the expressionof TNF-α and MDA were significantly lowered on scald shock with NaPyr groupat4h and8h postinjury(P<0.01).ConclusionsSodium pyruvate can significantly inhibit the increases of the kidneyvascular permeability in rats with scalded injury,and it would be helpful for theprevention of shock after scald and complication.
