ObjectiveTo investigate the apoptosis of bladder cancer cell5637induced by pseudo-laric acid B in vitro and further its mechanism.Aims to open up new plants inthe multimodal treatment of bladder cancer chemotherapy to provideexperimental data.MethodsThe cell proliferation was detected by MTT assay;the cell apoptosis wasobserved by inverted phase contrast microscope and flow cytometry withAnnexin V-FITC/PI double staining;the expressions of survivin protein andcaspase-3protein were detected by Western blot assay.ResultsIt showed that pseudolaric acid B remarkably induced apoptosis of5637cell line and the inhibition was depended on the exposure dose and time.IC50is165.91±20.75μmol·L-1、89.85±13.43μmol·L-1、48.31±2.54μmol·L-1when treatedwith PBA for24、48、72hours.Moreover, pseudolaric acid B suppressed survivinand up-regulated caspase-3expression.Conclusionspseudolaric acid B inhibits the proliferation and induces the apoptosis of5637cells.The molecular mechanism of pseudolaric acid B inducing theapoptosis of5637cells may be associated with its action of down-regulating theexpression of survivin,and up-regulating the expression of caspase-3.
