ObjectTo examine the effect of Pseudolaric acid B on the growth of human colon cancer cell line,sw620,and its possible mechanism of action.MethodColorectal carcinoma cell SW620 cultured in vitro was treated with various concentrations(0.5,1.0,2.0,4.0μmol/L)of PAB at various intervals. The inhibition of cell growth was examined by MTT assay.Cell cycle and apoptosis were analyzed by flow cytometry.ResultsPseudolaric acid B inhibited the growth of SW620 cells in a time-and dose-dependent manner.The inhibition ratio is 23 percent after the cells are treated with 0.5umol/L PAB for 24 hours,and the ratio is 74% after the cells are treated with 4 umol/L PAB for 72 hours.After the cells were treated with different concentration of PAB for 24h,the ratio of G0/G1 phase and S phase decreased,ratio of G2/M phase increased obviously.lt suggests that PAB could induce cell cycle arrest at the G2/M phase.CoclusionsPAB can dramatically suppress the SW620 cell growth.The possible mechanism of action is inducing apoptosis after G2/M phase arrest.
