ObjectiveTo investigate the apoptosis and the possible mechanism ofHuman osteosarcoma cell lines MG-63induced by Alpinetin.MethodAfter different concentration of Alpinetin treated for24hours invitro,the MG-63cell lines’ viability were assayed by MTT methods, cellcycle was analyzed by flow cytometry, cell apoptosis was detected byannexin V-FITC/PI double labeling, the expression of Caspase-3,Cytochrome C and Bcl-2were analyzed by Western blot.ResultAlpinetin could induce the apoptosis and suppress theproliferation of MG-63cell lines with the concentration dependedwhich were most apparently in80M. Meanwhile, Alpinetin couldaffect the cell cycle. Compared with control group, the80M groupdisplayed the increasing rate of apoptosis(P<0.01). The percentageof G0/G1increased and S+G2+M decreased (P<0.01). Caspase-3and Cytochrome C expressed increasingly and Bcl-2expresseddecreasingly (P<0.01). ConclusionsAlpinetin could induce the apoptosis and suppress theproliferation of Human osteosarcoma cell lines MG-63and affect thecell cycle, the mechanism were correlated with mitochondrial pathway.
