Abstract/Summary:
ObjectivesTo study the mechanism of Gancaofuzi Decoction onosteoporosis rats induced of PICP, ICTP, CathK and ColⅠ byovariectomy.Methods40SD rats were randomly divided into the sham operation group,ischemia-reperfusion group, the low and high dose groups of GCFZT andestrogen group,8rats in each group. Each rat had been record Ⅱelectrocardiographic (ECG) before the experiment. The sham operation groupwas only sham operated and the other four were ovariectomy. Then salinewere given to the sham operation group and ischemia-reperfusion group at10ml/kg weight; estrogen group were given to estrogen at2.7mg/kgweight; GCFZT were given to Low and High Groups at dose of12.5g/kg and50g/kg weight. All of these medicines were given to rats at10ml/kg weightand one time each day. The administration would continue3months. Until thethird month,The rats were killed and the sera were collect to determine themyocardium zymogram (PICP and ICTP); HE staining was employed toanalyze the pathological damage in myocardium of heart apex and andimmune staining to detect the protein expression of CathK and ColⅠ. Results1. Serum enzymes PICP and ICTP changes: Compared with the shamoperation group, the contents of ICTP increased significantly (P<0.01) andPICP decreased significantly in ischemia-reperfusion group; The contents ofICTP compared with ischemia-reperfusion group, the high and low groups ofGCFZT and isosorbide dinitrate group were decreased significantly (P<0.01).The contents of PICP compared with ischemia-reperfusion group, the highand low groups of GCFZT and isosorbide dinitrate group were increasedsignificantly2. Pathological changes: The sham operation group have significanthistological changes; Distal femur bone trabeculae number is more, thicknessis evener, form three-dimensional mesh structure. Compared with the shamoperation group,Rats trabecular bone number significantly reduce and thinner,the uneven thickness, span increase and shapes, stereo mesh structuredamage, bone marrow cavity expansion, the area of the note the amount of fatcells. Compared with the sham operation group,the osteoporosis of the highand low dose groups of GCFZT and Estradiol Benzoate group the number ofbony trabeculae were increasing, but low doses as high dose dose group ofobvious3. CathK and ColⅠ protein expression: Comparison with the shamoperation groups, expressions of CathK were increased and ColⅠ weredecreased, and variations were statistical significance (P<0.01); Comparedwith ischemia-reperfusion group, ColⅠ expressions of the high and low dosegroups of GCFZT and estrogen group were increased in varying degrees,CathK expression were decreased in varying degrees and were statisticallysignificant (P<0.01).ConclusionAccording the result of the experiment, We can conclude:GCFZT canprevent and curemechanism of postmenopause osteoporosis muti-targets. GCFZT possesses the double function of inhibition of bone absorbtion andacceleration of postmenopausal osteoporosis, also can decreas the proteinexpression of CathK and increas the protein expression of ColⅠ inhibit cellapoptosis.to accelerate bone cell multiplication and bone growth.
