ObjectiveStudy of caveolin-1and MMP-2expression in the lens epithelial cells fromoxidative damage cataract and to explore the effects of tea polyphenols onlens epithelial cells of CAV-1and MMP-2expression and to explore the TP mayprovide the experimental and theoretical basis of new drugs prevention and treatment ofcataracts.MethodsEstablish the experimental cataract model, set the untreated group,ultraviolet radiation group and experimental groups (namely ultravioletradiation+TP group), the experimental groups by tea polyphenolsconcentration is divided into four subgroups, namely0.02mg/L,0.2mg/L,2mg/L,20mg/L, respectively,6h,12h,24h,48h sampling,Morphological and histopathological observation.The expressions of CAV-1and MMP-2were detected by the S-P immunohist-ochemical method andWestern blot method in lens epithelial cells.ResultsBlank control group, rats were cultured48h in vitro lens to be transparent,UV irradiated groups of isolated rat lens with UV treatment time, the turbidity ofthe lens is gradually increased. Different concentrations of TP experimental group lens opacities progress more slowly than UV sets, in different timeperiods of turbidity were significantly lower than the UV group.The differencesof Caveolin-1and MMP-2expression between the experimental group and theUV group were statistically significant(P <0.05). At different times,CAV-1expression in the lens epithelial cells positive for the experimental group wassignificantly higher than the UV group, but MMP-2expression was significantlylower than the UV group,the degree of lens opacity for the experimental groupwas significantly lower than the UV group. When TP concentration is0.02μg/ml, the difference of Caveolin-1expression between the experimentalgroup and the untreated group were not statistically significant(P>0.05); thedifference of Caveolin-1expression between the rest of the experimentalgroup and the untreated group were statistically significant(P <0.05).ConclusionCAV-1exists in the normal lens epithelial cells, with the extension of the UVirradiation time, CAV-1expression gradually reduced; MMP-2was notexpressed in normal lens epithelial cells, with the extension of the UVirradiation time, the expression of MMP-2is gradually increased. TP can inhibitthe reduction in UV-induced lens epithelial cell of CAV-1expression reduceand MMP-2increase, which, when tea polyphenols concentration is0.02mg/L,the effect is the best.
