ObjectiveIn recent years a large number of studies have shown that the injury inendothelial cell play a catalytic role in the formation and development of coronaryheart disease (CHD).Endothelial progenitor cells (EPCs) are precursor cells ofendothelial cells. EPCs participate in the repair process of endothelial injury,adiponectin (APN) affect the number and function of EPCs. Directly or indirectly toenhance the repair capacity of endothelial injury, and promote the recovery of thenumber and function of endothelial cells, thereby delaying the occurrence anddevelopment of lesions AS.This article is analyzed by enzyme-linkedimmunosorbent assay (ELISA), and by flow cytometry method for thedetermination of coronary artery disease in peripheral blood of patients with APN,the level of CD34~+/CD309~+EPCs, and to analyze the trend of changes in theseverity of the disease. Select normal as controls to observe the size of coronaryheart disease in the vascular endothelial injury.By giving oral lipid-lowering drugswere followed up for two weeks, the determination of the above factors, toinvestigate the APN and EPCs number and function to restore the role oflipid-lowering drugs in addition to the Lipid Metabolism.MethodsChose patients first diagnosed120cases of coronary heart disease andsuspected coronary artery disease by percutaneous transluminal coronaryangiography (CAG) confirmed coronary heart disease patients at the Department of Cardiology of our hospital from October2011to March2012, including acutecoronary syndrome45cases,51cases of stable angina pectoris, and select the24cases of healthy control group; ACS group were admitted to hospitalimmediately, SAP group admitted to hospital the next morning, fasting venousblood examination and blood glucose, lipids and other basic indicators;Extractof fresh heparin lithium as anticoagulant of fasting blood samples100μl wereadded to centrifuge tubes labeled A, B, tube, take the A tube as negative control,tube B by adding10μl of FTIC-labeled CD34antibody and10μl of the PElabeled CD309monoclonal antibody flow cytometry assay of CD34~+/CD309~+EPCs, the proportion of whole blood;Specimens of the above-mentioned anti-clotting to take2ml instantly at room temperature for30min,3000r/mincentrifugation10-15min plasma stored at-80℃refrigerator, measuredadiponectin level by ELISA; given on all patients with coronary artery disease,Swiss Shu atorvastatin calcium medication (10mg1/late) intervention,outpatient follow-up after two weeks, again using ELISA method detectedadiponectin levels by flow cytometry in peripheral blood of patients with CD34~+/CD309~+EPCs, the proportion of whole blood,to observatie f lipid-lowering drugsto improve the terms of the value of the APN EPCs number and function.Results(1) Baseline data of the study groups in age, sex, weight, smoking, bloodlipids, blood pressure, creatinine, urea nitrogen, fasting blood glucose and otherrisk factors there was no significant difference (P>0.05).(2)Before the intervention of application Ruishu cutting Ting calciumlipid-lowering drugs, ACS peripheral blood adiponectin and CD34~+/CD309~+EPCs,were lower than that in SAP and control group, difference statisticallysignificant (P <0.01); SAP and control group have also difference statisticallysignificant (P <0.05).(3)With coronary artery lesions progressively heavier both showed adownward trend, suggesting that hypoadiponectinemia and endothelial progenitorcells in peripheral blood level decreased easy to make coronary diffuse diseaseoccurs.(4)The level of correlation analysis showed that adiponectin and CD34~+/ CD309~+EPCs were positively correlated (r=0.865,P﹤0.01).(5)Compared with before the intervention, total cholesterol, LDL cholesterolshowed a significantly decreased, the difference was significant (P <0.01),triglycerides showed a downward leavel,difference significant statistically (P<0.05), while the high-density lipoprotein cholesterol changes was statisticallysignificant (P>0.05). Adiponectin and CD34~+/CD309~+EPCs, level than beforetreatment significantly increased (P <0.05).ConclusionsVascular endothelial injury leads to the functional state of disorder, andvarious types of cardiovascular disease is closely related to this,to develop theprevention and treatment of cardiovascular disease strategy,we should first repairthe damage of vascular endothelial and start endothelialization.This study foundthat the levels of APN and CD34~+/CD309~+EPCs in the CHD group comparedwith the healthy group was significantly decreased and increased with the severityand the cumulative range, the more decline in its value;by lipid-lowering drugsafter the intervention, the factor levels than medication previous significantincrease.In summary,for patients with coronary heart disease, not only to get alipid-lowering and more the importance is to protect the endothelial cells,increasing the content of adiponectin, reduce endothelial damage, and ultimatelydelay the progress of atherosclerosis.
