OBJECTIVE:The rhizome of Polygonatum. Kongianum, P. sibiricum, P. cyrtonema is a kind of Chinese traditional drug accepted by the Pharmacopoeia of China. But many kinds of plants which belong to Polygonatum.were used as it. The character of Rhizoma Polygonati is similar to its fakes, but their pharmacological effect is quite different. So it is necessary to set up an effective method for the identification of them. This study tends to set up a new method for the identification of Rhizoma Polygonati and its fakes such as Polygonatum odoratum, P.cirrhifolium, P. macropodium, P. humile, and P. filipes.The. new method was developed for identification of Chinese traditional medicine Rhizoma Polygonati from its fakes. The scavenging effect of the traditional Chinese medicine extracts on superoxide anion free-radical was studied in detail. METHODS:The traditional methods and the DNA molecular mark technology were used for identification of Rhizoma Polygonati and its fakes which were collected from Jiangxi Province, Guizhou Province, Beijing, BaXian Mountain, Wuling Mountain, Jiu Shan Ding Mountain.28arbitrary premiers were evaluated for the analysis of RAPD(random amplified polymorphic DNA)、ISSR(inter-simple sequence repeat), The superoxide free-radical was produced by the autoxidation of pyrogallol in alkaline solution and the scavenging effects of Rhizoma Polygonati and the Chinese traditional medicines extracts were detected by single sweep oxcillopolarograpgy. RESULTS:It is very difficult to identify Rhizoma Polygonati from its fakes by traditional identification but DNA molecular mark technology such as ISSR and RAPD can solve this problem.28arbitrary primers were evaluated and some of them such as TGCCGAGCTG, GAAACGGGTG, GTGTGCCCCA, CACCGTATCC,(AC)8T,(ATG)5,(CA)8G,(TC)8G,(AG)8T, CTGGGGCTGA, GCTGGCTGAC, TCCCGAACCG were selected for the study of the isolates.After PCR, all the mixture was separated by1.8%agarose gel. The genetic diversity among7species of Polygonatum, including different area species, was investigated by DNA molecular mark technology. The systemic cluster analysis indicated the relationship between the accessions of Polygonatum. Single sweep oxcillopolarograpgy was simple to detect the scavenging effect of the traditional Chinese medicine extracts on superoxide anion free-radical.CONCLUSION:The proposed method can identify Rhizoma Polygonati from its fakes such as Polygonatum odoratum, P.cirrhifolium, P. macropodium, P. humile, and P. filipes. It is a rapid, effective method and could be a criterion for the identification of the plants of Polygonatum. The different capability of scavenging effect on superoxide anion radicals was shown for TCM processed by different methods.
