Objecive: Lectin-like oxidized low density lipoprotein receptor-1(LOX-1)is the recepter of ox-LDL which plays an important role in thedevelopment of Atherosclerosis (AS).The purpose of this experiment is tostudy the relationship between the lesion degree of coronary artery diseaseand the level of serum lox-1、hs-CRP as well as the LOX-1mRNA in humanperipheral blood mononuclear cells.Methods:138consecutive patients who accepted newly coronaryangiographyin Affiliated Hospital of North Sichuan Medical College during2011.11to2012.11were collected.According to the result of coronaryangiography,the patients were divided into study group (106cases) who werediagnosed with coronary heart disease while the control group (32personswhich no suffering from coronary heart disease).In the study group,Theirangiograms were scored according to the SYNATX score. The patients weredivided into tertiles according to the Syntax score: lowest Syntax score tertile(Syntax score22;37cases), intermediate SYNTAX score tertile (Syntaxscore of23to32;33cases), and the highest Syntax score tertile (Syntax score33;36cases).The LOX-1, hs-CRP protein expression in serum and LOX-1mRNA expression in peripheral blood mononuclear cells were detected byELISA and qRT-PCR assay respectively to explore the relationship betweenthe level of LOX-1, hs-CRP and the severity of coronary artery disease. Results:1、General clinical data shows the differences of sex, age,history of hypertension、diabetes mellitus、smoking as well as the levels ofTG、V-LDL were not significant between study group and control group(P>0.05),while the levels of TC、LDL-C were statistically significantbetween two groups(P<0.05);in the study group,the levels of TC、LDL-Cwere statistically significant among tertiles(P<0.05),the differences of agewas higher in the highest Syntax score tertile than other tertiles(P<0.05).2、the levels of LOX-1、hs-CRP、LOX-1mRNA in study group werehigher than the control group(113.641±5.818pg/ml vs104.711±14.850pg/ml;2.096±0.663mg/l vs1.482±0.511mg/l;0.362±0.071vs0.286±0.077;P<0.01respectively)。3、in the study group,the levels LOX-1were significantlydecreased among patients in the lowest Syntax score tertile(108.967±4.000pg/ml) as compared with those with the intermediateSYNTAX score tertile (113.694±5.125pg/ml) or highest SYNTAX scoretertile (118.398±3.829pg/ml) as well as the leves of hs-CRP(1.788±0.623mg/l vs2.095±0.611mg/l vs2.412±0.614mg/l,P<0.05);thedifference of LOX-1mRNA expression on PBMC were statistically significantin the lowest SYNTAX score tertile as compared with those with theintermediate SYNTAX score tertile or highest SYNTAX scoretertile(0.318±0.450vs0.421±0.069vs0.349±0.057, P<0.05).4、Correlation analysis:There was positive correlation between thelevel of hs-CRP、LOX-1、LOX-1mRNA and the SYNTAX scroe(P<0.05). Multiple stepwise regression analysis showed that the LOX-1wasthe independent impact factors of hs-CRP(t=3.560,P<0.01);TC and LD L-C were the Independent impact factors of LOX-1mRNA(t=2.394,t=0.205,P<0.05);LOX-1and LOX-1mRNA were theindependent impact factors of the SYNTAX score.(t=5.930,t=4.040,P<0.01)。Conclusion: The level of LOX-1、hs-CRP and the expression of LOX-1mRNA on PBMC in patients were increased with the severity of coronaryheart disease,which suggest those factors may be involved in thedevelopment of coronary atherosclerosis.Combined detection of plasmaLOX-1, hs-CRP and LOX-1mRNA expression on peripheral bloodmononuclear cells can be used as a new indicator of the degree of coronaryartery disease.
