Objective:Detection the expression of Annexin A1in acinetobacter baumannii sepsis rats in different time point of sepsis, and treatment the groups by xuebijing injiection, through testing the related inflammation factor TNF-αand IL-8, Discussing the relationship between Annexin Al and acinetobacter baumannii sepsis and the effect of xuebijing injiection to Annexin Al and acinetobacter baumannii sepsis.Methods:Using54healthy wistar rats were randomly divided into normal control group (N group), the sepsis group (B group) and treatment group (X group), N group inject by physiological saline into enterocoelia, B group inject by acinetobacter baumannii (ATCC19606) into enterocoelia, X group inject by acinetobacter baumannii and xuebijing injection at the same time. Each group is divided into6h,12h,24h three subgroups, each subgroup6rats. Then,picking up every rat,s arterial blood neutrophils, through Western blot detect the expression of Annexin Al in neutrophils, the expression of Annexin A1in rat lungs through immunohistochemical EliVisionTM plus, separation serum of rat and detection serum IL-8and TNF-αby ELISA method.Results:1. Successfully establish normal control group, the sepsis group and the treatment group rats model.2.Levels of Annexin Al protein in rat neutrophils in the experimental design of three observation time points (6h,12h,24h):in sepsis group protein levels were significantly elevated to the normal group, the difference was statistically significant (p<0.05); treatment group protein increased significantly to the sepsis group, the difference was statistically significant (p<0.05). Sepsis6h group higher than normal group, the difference was statistically significant (p<0.05); Sepsis24h and sepsis12 h group up from sepsis6groups, the difference was statistically significant (p<0.05); Sepsis12h and sepsis24h has no statistical significance (p>0.05) difference.3.Levels of IL-8in rat serum in the experimental design of three observation time points (6h,12h,24h):in sepsis group IL-8levels were significantly elevated to the normal group, the difference was statistically significant (p<0.05);levels of treatment group reduce significantly to the sepsis group, the difference was statistically significant (p<0.05). Sepsis6h group higher than normal group, the difference was statistically significant (p<0.05); Sepsis24h and sepsis12h group up from sepsis6groups, the difference was statistically significant (p<0.05); Sepsis12h and sepsis24h has no statistical significance (p>0.05) difference.4.Levels of TNF-ain rat serum in the experimental design of three observation time points (6h,12h,24h):in sepsis group TNF-a levels were significantly elevated to the normal group, the difference was statistically significant (p<0.05);levels of treatment group reduce significantly to the sepsis group, the difference was statistically significant (p<0.05). In sepsis group Annexin Al expression are higher than normal lung tissue, compared with normal group, with statistical difference (p<0.05); the expression ofAnnexin Al in treatment group (6h,12h,24h) partial rendering strong positive expression, compared with sepsis group was statistically difference (p<0.05).Conclusion:1.Levels of Annexin Al in sepsis rats neutrophils and lungs, with the development of disease increased, and reach a balance between12h to24h; the intervention of injecting xuebijing after sepsis in rats can improve the level of Annexin Al in rats.2.The levels of TNF-aand IL-8in sepsis rats serum are increased with the development of disease; the intervention of injecting xuebijing can reduce the levels of TNF-a and IL-8in serum. 3.Xuebijing injection can effectively be helpful to the acinetobacter baumannii sepsis rats, significantly inhibiting the levels of LI-8and TNF-a;it Can improve the Annexin A1content in the body that play the role of anti-inflammatory at the same time, but how Annexin A1protein inside the body to affect the level of IL8and TNF-a and specific mechanism remains to be further confirmed.
