Objective:Endometrial carcinoma occurs in endometrial cancer malignant tumors, is one of the most common tumors in female reproductive system, and is the result of third malignant gynecological tumor death. With the development of society and the improvement of economic conditions, endometrial cancer incidence increased year by year,being the upward trend in our country. Cell fate determination factor Dachshund(DACH1) is a mammalian homologue gene of Drosophila dachshund, by1which down-regulation of epidermal growth factor(EGFR) and cyclin D1way to inhibit the proliferation of tumor cells, that have been reported in human breast cancer, ovarian cancer, thyroid papillary carcinoma, prostate cancer is a tumor suppressor gene. Fatty acid synthase (FASN) is the key enzyme involved in tumor formation of fat, which is a common expression in cancer. Sterol element binding protein (sterol regulatory element binding proteinl, SREBP1) can regulate a variety of genes involved in lipid synthesis.one subtype SREBP1-c is connected to the adjusting element of FASN promoter, which can increase the expression of FASN transcription. Therefore, in this study, immunohistochemical SP method was used to detect expression situation of DACH1, FASN, SREBP1in endometrial carcinoma, atypical hyperplasia endometrium, endometrial, complexity of normal endometrium, to explore the relationship with endometrial cancer clinical pathological parametersMethods:Immunohistochemical staining was used to detect the expression of DACHl protein, FASN, SREBP1in20cases of normal endometrium,10cases of complex endometrial hyperplasia,10cases of atypical endometrial hyperplasia and60cases of endometrial carcinoma. Using x2to test the relationship between the expression of DACH1protein, FASN, SREBP1and the different clinical pathological features. Using Breslow(Generalized Wilcoxon) survival analysis to compare the expression of three and prognosis.Results:(1)The expression of DACH1protein in endometrium carcinoma, atypical hyperplasia endometrium complexity of endometrial hyperplasia and normal endometrium was increased, the difference was statistically significant (P<0.05);FASN and SREBP1were decreased in endometrium carcinoma, atypical hyperplasia endometrium complexity of endometrial hyperplasia and normal endometrium, the differences were statistical significance (P<0.05);(2)The expression of DACH1protein in endometrial carcinoma was related with the degree of differentiation,the operation-pathological staging and the pathologic type (P<0.05), and the expression of DACH1protein in the higher differentiation, the earlier clinical stage and type I in endometrial carcinoma which was more obvious; The expression of FASN was associated with endometrial carcinoma differentiation (P<0.05),which in the lower differentiation was higher; The expression of SREBP1was associated with endometrial differentiation and age (P<0.05). which in lower differentiation and the older was more obvious;(3)The expression of DACH1protein was negatively correlated with the expression of FASN, SREBP1in endometrial carcinoma(r=-0.41, P<0.05; r=-0.40, P<0.05); The expression of FASN was positively correlated with the expression of SREBP1(r=0.86,P<0.05);(4)In endometrial carcinoma,the overall survival rate of DACH1protein positive patients was significantly higher than DACH1protein negative patients(P<0.05); Compared with FASN negative group, the overall survival rate of FASN positive group was significantly lower(P<0.05); Compared with SREBP1negative group, the overall survival rate of SREBP1positive group was significantly lower(P<0.05).Conclusion:The absence or low expression of DACH1and the overexpression of FASN and SREBP1means the higher invasiveness and poor prognosis in endometrial carcinoma, above three kinds of protein expression in endometrial carcinoma have correlations with each other, there is a certain relationship with the progression of endometrial carcinoma.
