Background:Stroke is one of the common reasons of death and being maimed, there will be one patient died in each21seconds on China mainland on average from stroke, and there will be a new case in each12seconds. The number of this classic patients was up to7.49million currently,but the frequent of this disease is still high, it has became the second cause of death and first cause of being maimed of Chinese diseases. About two-thirds of the existing patients needed nursing care, this is a heavy burdens on individuals、families、community and so on. One class of stroke:ischemic stroke, which accounts for the main part, for a long time, has been lack of effective means of defendent and treatment, most patients are left with vary degrees of paralysis, pain, and cognitive disorders. Although various scholars was probing the pathophysiological mechanisms of stroke, there has not been any development to promote the recovery of neurological function after stroke patients so far; there has not been any development of effective medicines and other treatments, which makes things more worse is that most studies are still in the theoretical stage.Objective:To study the stability of the MCAO operation and the neural processes of reproduction after reperfusion of MCAO rats; To probe the expression of Wnt7b and the main molecules of its classical signaling pathway:β-catenin in hippocampal dentate gyrus in order to make suer of the function of Wnt7b’ in the process of nerve regeneration after ischemia-reperfusion.Methods:60Wistar rats were assigned to contrary group and focal cerebral ischemia-reperfusion group randomly, the operation groups were reperfused after90min. Neurological function were scored on2h,1d,3d,7d,14d., taking out the hippocampus in the brain tissue of rats on each of those points, different groups were detected by RT-PCR, Q-PCR, immunofluorescence techniques to determine the expression of Wnt7b, the expression of β-catenin was probed by using immunofluorescence techniques in rat after nerve injury to explore Wnt7b’ role and mechanisms in nerve regeneration.Results:Thread occlusion method can establish cerebral ischemia-reperfusion model in rats stabilitly;, the scores of adult Wistar rats after cerebral ischemia-reperfusion on2h is higher than normal nerve function group (P<0.01), as well as the groups of3d,7d,14d(P<0.05); The expression of Wnt7b in normal adult rats is lower than the injury ones, during the process of nerve regeneration, Wnt7b mRNA increased significantly(P<0.01):The important molecules of classic signaling pathway:β--catenin also significantly increased along with the expression of Wnt7b (P<0.01).Conclusion:90min middle cerebral artery occlusion (MCAO) in rats can build a simple stability ideal model of cerebral ischemia-reperfusion injury, it is suitable for broad application in the basic experimental research;Such damage can stimulate short regeneration of neural stem cells in the SGZ; Wnt7b mRNA、Wnt7b protein and β-catenin were approved higher than normals in our experiment firstly by Q-PCR、 RT-PCR、immune fluorescent. This topic explains that Wnt7b may effect the process of regeneration through classic Wnt/β-catenin signaling pathway, which provides the theory of further experiment:probing the effect of upregulating or downregulating the quantity of Wnt7b and provides a new way of stem cells therapy.
