Objective: To study the mRNA level and protein expression of Beclin1and Atg5inovarian epithelial cancer and its significance.Methods: Detect the mRNA content and protein expression of Beclin1and Atg5ofhuman in30cases of epithelial ovarian cancer tissue,30cases of benign ovarian tumortissue and30cases of normal ovarian tissue by immunohistochemical staining and ReverseTranscriptase Polymerase Chain Reaction (RT-PCR) semi-quantitative method,respectivel-y. Analyze the relationship between Beclin1and Atg5in epithelial ovarian cancer tissues,and the relationship between Beclin1and Atg5with clinical pathological features such asclinical stage and cell differentiation of epithelial ovarian cancer.Results:1. The mean mRNA level of Beclin1was0.2431in epithelial ovarian cancer tissue,0.6154in benign ovarian tumor tissue and0.6951in normal ovarian tissue. The positiverate of Beclin1protein expression was36.7%(11/30) in epithelial ovarian cancer tissue,70%(21/30) in benign ovarian tumor tissue and83.3%(25/30) in normal ovarian tissue.There were no statistical difference in benign ovarian tumor tissue and normal ovariantissue (P>0.05), but the mRNA level of Beclin1and the positive rate of Beclin1proteinwas significantly decreased in epithelial ovarian cancer tissue compared to that in the othergroups (P<0.05).2. The mean mRNA level of Atg5was0.3002in epithelial ovarian cancer tissue,0.5235in benign ovarian tumor tissue and0.6244in normal ovarian tissue. The positiverate of Atg5protein expression was33.3%(10/30) in epithelial ovarian cancer tissue,60%(18/30) in benign ovarian tumor tissue and73.3%(22/30) in normal ovarian tissue. Therewere no statistical difference in benign ovarian tumor tissue and normal ovarian tissue (P >0.05), but the mRNA level of Atg5and the positive rate of Atg5protein was significant-ly decreased in epithelial ovarian cancer tissue compared to that in the other groups (P<0.05).3. The mRNA level and positive expression rates of Beclin1and Atg5proteins inovarian epithelial cancer was correlated with clinical stage and pathological grade (P<0.05), but have no significant correlation with the age, the size of solid tumors and whetherthere is ascites、lymph node metastasis (P>0.05).4. In ovarian epithelial cancer,the expression of Beclin1was not correlated with Atg5(r=0.049, P=0.548).5. The expression of Beclin1、Atg5in different histological types of ovarian epithelialcancer have no statistical difference (P>0.05).Conclusions:1. Expressions of Beclin1and Atg5were down regulated in epithelial ovarian cancertissues.The inhibition of autophagic capacity may be related to tumorigenesis and thedevelopment of epithelial ovarian cancer.2. The expression of Beclin1was not correlated with Atg5.3. The expression of Beclin1、Atg5in different histological types of ovarian epithelialcancer have no statistical difference.4. The detection of Beclin1and Atg5in mRNA and protein expression levels may beconsidered as important indicators of determine the type of epithelial ovarian tumors andearly diagnosis and treatment of ovarian cancer patients,observe the mechanism of autoph-agy can provide a new and effective therapeutic schedule for the treatment of epithelialovarian cancer.
