Objective Allograft was constructed by decellularized muscle bioscaffolds (DMBS) seeded with bone mesenchymal stem cells (BMSCs),this trial aimed to investigate the effects of allogenic transplantation on repairing acute hemi-trancetion SCI in rats.Methods DMBS were prepared by reformed chemical approach and sterilized by compound cold sterilization, BMSCs were harvested by density gradient centrifugation and adherent-cultured. The third generation cells were injected into DMBS to construct the BMSCs-DMBS Allograft, which biocompatibility was observed under SEM and fluorescence microscope in vitro at2weeks,48adult female SD rats were subjected hemi-transect at T10level to build SCI model then divided randomly into4groups (each group, n=12), BMSCs-DMBS, BMSCs and DMBS were implanted in groups A, B, and C, respectively. Group D was blank control by inject PBS. At1,2,3and4weeks after surgery, the functional recovery of the hind limbs was evaluated by the Basso-Beattie-Bresnahan (BBB) locomotor rating score,At4weeks after surgery, HE staining and immunofluorescence assay were adopted.Results Masson staining showed the internal structure of DMBS in parallel rows,which mainly were collagenous fibers without muscle fibers. BMSCs were injected into scaffolds and cultivated for14days, abundance surviving cells labeled by Hoechest33342, which been confirmed flourished adhering to the scaffolds by SEM, Hematoxyllin-eosin staining revealed that the cavity was smaller in group A than other there groups, immunofluorescence showed that more neurofilament200(NF-200) positive fibers and Nestin positive cells were detected in the lesioned area in group A than that in group B,C and D, but significantly less GFAP positive cells. The transplanted BMSCs labeled by hoechest33342were migrated into anterior angle of contralateral gray matter and some morphologically simulated the neurons. The BBB scores at2,3and4weeks after surgery in group A were significantly higher than those in other3groups, the differences were statistically significant (P<0.01),at4weeks group B was higher than group C, the differences were statistically significant (P<0.05),group D was the lowest at each test time(P <0.01).Conclusion With relatively regular internal structures, DMBS which can be biocompatible with BMSCs,inhibit glial scar and enhance the survival, migration and differentiation of BMSCs,are the ideal nature vector for cell transplantation. Synergistic effect of co-transplant DMBS with BMSCs can promote rat motor function recover.
