ObjectiveSpontaneous abortion is occurred in the natural state (non-human factors),accounting for10%-15%of all pregnancy.It brings to the woman not only physical pain but also psychological pain. While, its etiology is complex and the pathogenesis is unclear. Recently, more and more scholars began to focus on the relationship between oxidative stress and spontaneous abortion. Studies have been shown that human embryonic developed under hypoxic environment. Trophoblast cells which grow in low maternal blood flow is very sensitive to oxidative stress. When the maternal blood flow into the villus gap, trophoblast cells will transform from hypoperfusion and hypoxia into high perfusion and high-oxygen environment,and large amounts of reactive oxygen species (ROS) will be produced. In order to adapt to this change, the body need to generate the corresponding antioxidants. If antioxidant defense is not enough to eliminate ROS, the accumulation of ROS will produce oxidative stress. The cell will be injured by lipid peroxidation, DNA oxidative damage and DNA strand breaks, leading to pregnancy failure. Previous studies show that oxidative stress markers were significantly increased in early pregnancy failure villus, suggesting that the oxidant/antioxidant imbalance and tend to oxidative stress. The main antioxidant gene includes PTEN and Prx2in early pregnancy women villus. PTEN play an important role in embryo development and implantation, cell migration, and apoptosis. The Prx2is an important antioxidant defense protein, and regulated by PTEN.Study about the expression and significance of PTEN and Prx2in spontaneous abortion vills has not been reported at domestic and abroad.From this paper, Immunohistochemistry SP and RT-PCR method will be used to detect the PTEN and Prx2protein and mRNA expression. We can try to find out of the mechanism of PTEN and Prx2play in spontaneous abortion In the field of oxidative stress.MethodWe use immunohistochemistry SP and RT-PCR method to detect the PTEN and Prx2protein and mRNA expression in experimental group and control group. Data will be analyzed by the SPSS17.0software system. P<0.05is statistically significant.Result(1) PTEN protein expression PTEN protein expression in the nucleus of trophoblast cells. The expression of protein of PTEN in experimental group(70%) is significantly lower than that of control group(93.3%).Corrected=4.007, P=0.045, P<0.05. The difference is statistically significant.(2) Prx2protein expression Prx2protein expression in the Cytoplasm of trophoblast cells. The expression of protein of Prx2in experimental group(36.7%) is significantly lower than that of control group(76.6%).Rank sum test H=25.195, P<0.001, P<0.05. The difference is statistically significant.(3) PTEN mRNA expression PTEN mRNA expression in experimental group(0.059±0.010) is significantly lower than that of control group (0.929±0.100), Correction t-test t=47.339, P<0.001.The difference is statistically significant. The result is Consistent with the immunohistochemistry analysis of the experiment.(4) Prx2mRNA expression Prx2mRNA expression in experimental group(0.222±0.029) is significantly lower than that of control group (1.131±0.125), Correction t-test t=38.847, P<0.001.The difference is statistically significant. The result is Consistent with the immunohistochemistry analysis of the experiment.(5) Correlation Analysis PTEN and Prx2are positively correlated, r=0.615, P<0.001.Conclusion1. PTEN and Prx2are important antioxidant system in body. That the reduced expression in spontaneous abortion of the two gene is likely to have a certain relationship with the occurrence of spontaneous abortion.2. PTEN and Prx2in patients of spontaneous abortion villus are positively correlated. It suggesting that the two gene may play a synergistic role in the pathogenesis of spontaneous abortion.
