Objective: To detect the nitric oxide (NO) concentration, nitric oxide synthase (NOS) activity, vascular endothelium growth factor (VEGF) concentration in serum and NO concentration, NOS activity, p53 protein and Bcl-2 protein expression in chorionic villus collected from the patients with recurrent spontaneous abortion(RSA) and the normal pregnancy artificial abortion, to compare the potential difference between the two groups and in order to highlight the possible pathogenesis of RSA.Methods: In this study, 33 cases of RSA and 30 cases of normal pregnancy artificial abortion groups (control group) were examined. NO concentration, NOS activity in serum and chorionic villus were measured by colorimetry. VEGF concentration in serum was examined using the method of ELISA. p53 protein and Bcl-2 protein expression were measured by flow cytometer. The statistical data was analysed with Independent-Samples T Test and Pearson Correlation Analysis.Results: 1. The concentration of NO in serum for RSA group and control group were 77.21 + 30.78( mol/L) & 51.78 + 27.04(u mol/L), respectively. The NO concentration of RSA group was higher than that of control group, the difference was obviously (p<0.01). The concentration of NO in chorionic villus for RSA group and control group were 5.86 + 1.84( ti mol/g Protein) and 4.38 1.12( mol/g Protein), respectively.The NO concentration of RSA group was higher than that of control group, the difference also showed statistic significance (p<0.01). 2. The activity of NOS in serum of RSA group and control group were 29.51 3.17 (U/ml) & 25.78 + 3.82 (U/ml), respectively. The NOS activity in RSA group was higher than that of control group, the prominent difference was shown (p<0.01). The activity of NOS in chorionic villus in RSA group and control group were 5.74+1.49(U/mg Protein) and 4.42 0.80(U/mg Protein), respectively. The NOS activity of RSA group was statistically higher than that of control group (p<0.01).3. The concentration of VEGF in serum in RSA group and control group were 103.48 17.70(pg/ml) & 87.17 18.70(pg/ml), respectively. The VEGF concentration of RSA group was higher than that of control group, the difference with statistic significance was seen (p<0.01). 4. The expression of p53 protein in chorionic villus in RSA group and control group were 1.33 + 0.06 & 1.19 + 0.06, respectively, showing that the p53 protein expression of RSA group was obviously higher than that of control group (p<0.01). 5. The expression of Bcl-2 protein in chorionic villus in RSA group and control group were 1.27 0.08 & 1.63+0.15, respectively. The Bcl-2 protein expression of the RSA was lower than that of control group, showing statistic significance (p<0.01).Conclusions: NO concentration, NOS activity, VEGF concentration in serum and NO concentration, NOS activity, p53 protein expression in chorionic villus of RSA group were distinctly increased than that of control group, but Bcl-2 protein expression in chorionic villus was obviously decreased than that of control group, which would likely lead to more embryo cells prone to occur apoptosis and be responsible for RSA.
