Background and ObjectivesChemotherapy is one of the main means for treatment of cancer and it can lead to a series of side effects. There is no choice to injure normal tissue cells and tumour cells. All of them, myelosuppression is very commom. When it is serious enough chemotherapy could not go on as schedule so that may result in chemotherapy failure. How to reduce myelosuppression is important for the success of chemotherapy. So it is necessary for seeking effective methods to reduce myelosuppression. Now, there are some drugs for treatment myelosuppression caused by chemotherapy. For example, Leucogen, vitamin B4etc, the curative effect of them are not good relatively. The other ways are hematopoietic stem cell transplantation or blood component transfusion, but both of which are hard to use repeatedly and extensively. Newer drugs for elevating leukocytes like recombinant human granulocyte colony-stimulating factor(G-CSF) granulocyte-macrophage colony-stimulating factor (GM-CSF) etc. still appear limited effectiveness, high price and some side effects. Therefore exploring new drugs or methodsTangcaopian is the first SFDA approved drug for treatment of AIDS with pure traditional Chinese medicine including20species of medicinal plants. Geranium, honeysuckle, bupleurum, astragalus, licorice, Caulis Spatholobi, Oldenlandia diffusa, Ginkgo biloba are the main ingredients, which could clear away heat and toxic material, promote blood circulation and remove blood stasis, eliminate pathogen centralizer ect. It can improve symptoms such as inappetence, fatigue, diarrhea, lose hair, etc. Clinical results show that patients with long term administration of the drug could enhance their antiviral ability, improve life quality, delay the progress of the disease and reduce side effects of Western medicine. Most importantly it can increase patient’s CD4+lymphocyte number and CD4+/CD8+ratio. However the mechanism is not clear. The purpose of this study is to test whether Tangcaopian has the ability to accelerate the recovery of myelosuppressed mice or to protect bone marrow from chemotherapy drugs.Methods(1) Experimental groups and dosage:sixty mice were randomly divided into6groups including normal control group, model group, the model+Tangcaopian (low, medium, high dose groups with0.5,1.0,2.0g/kg body weight respectively), positive control group. Tangcaopian was given i.g. for21days.5Days later from the beginning of administration Tangcaopian intraperitoneal injection of mitomycin3g·kg-1was carried out and2g·kg-1at the10th day. Positive control group after the model successfully built was given G-CSF with hypodermic injection50μg/kg. Determining peripheral blood numbers at different time. At the end of the experiment, bone marrow cell counting and colony-forming cell culture were carried out.(2) a tumor-burdened animal experiment grouping and dosage:animals were randomly divided into5groups according to the weight,6in each group. Divided into normal control group, a tumor-burdened group, a tumor-burdened animal+tang grass tablets (1.0g/kg body weight) in the dose group, a tumor-burdened animal+tang grass tablets (1.0g/kg body weight) in the dose group+mitomycin (5.0mg/kg body weight) group, a tumor-burdened animal+mitomycin (5.0mg/kg body weight). Prevent application tang grass piece for five days, respectively to vaccinate mice S180sarcoma in mice and ascites tumor (0.2ml)/107, and the next day at the same time use chemotherapy drugs mitomycin5.0mg/kg treatment again. Tang grass continuous dosing, a total of18days. Determination of the following values, the tail vein of mice blood blood cell analyzer, respectively, the count number of peripheral blood white blood cells (WBC), platelet (PLT) count, the number of red blood cells (RBC), hemoglobin (Hgb) content, etc. Respectively at different time after dosing counting cells (or peripheral blood white blood cell count fell to terminate count); Femoral nucleated cell count:at the end of the experiment executed by Ann dead mice, on one side of the femur, conventional rushed out of the bone marrow nucleated cells, blood cells counter count number of nucleated cells. Cells in bone marrow CFU GM:at the end of the experiment executed by Ann dead mice, take a side femoral bone marrow cells, made from6orifice methyl cellulose, each sample set3hole, counting grains-giant cell colony count/105nucleated cells. Weigh weight in mice; A tumor-burdened animals were observed YiLiuLv (solid tumor) and animals live longer (ascites tumor), and the determination of peripheral blood cell count, etc. Experimental data with mean and standard deviation (s) X said that using SPSS10.0statistical analysis software for experimental data analysis, by comparing multiple samples mean of statistical analysis, single factor variance analysis take P<0.05level of significance test.Results(1)To the change of peripheral blood leukocyte count,18days after the first injection of mitomycin (model built on the eighth day), groups of white blood cell count recovered, but significantly lower than the control group;24days, tangcaopian in the white blood cell count and positive control group, low dose group was obviously higher than the model control group (p<0.05), up to the level of the normal control group;33days treatment group remained at a higher level of normal control group, model control group was lower than normal (p<0.05).40days, tang grass in dosage group and positive control group of white blood cells has started to fall, tang grass piece still maintain at a high level in the lower dosage treatment groups.(2) The number of neutrophils and lymphocytes change:the tendency of the number of neutrophils and lymphocytes and peripheral blood WBC count is broadly in line with a trend, are pieces of tang grass elevated low dose group showed better effect.(3) The number of peripheral blood erythrocyte changes:24days before after chemotherapy, the treatment group red blood cell count increases are not obvious, even lower than normal control group, but33days later, tang grass low dose group of red blood cell count is higher than the model group, continue to the end of the experiment, the results suggest tang grass tablets in the role of hematopoietic cells are obvious.(4) The number of bone marrow cells changes:groups of bone marrow nucleated cell count difference is not big, tang grass piece of low dose group of them have showed a downward trend, may number is the group of cells has reached normal levels, the body’s own adjustment has been caused by inhibition of bone marrow hematopoietic function.(5) Animals weight:weight in10days after chemotherapy, is generally lower than normal control group, but compared to before chemotherapy did not significantly reduce,(6) Tangcaopian tablets in mitomycin antitumor effects:tangcaopian group and treatment group, tumors), middle watery liquid, mitomycin and joint group small tumors, no liquid water in the middle. Tangcaopian piece and mitomycin alone have no obvious inhibition effect on S180sarcoma, but united both in animals inhibition rate of43.5%. For vaccination of ascites tumor mitomycin43days separate application group an animal ascites and death, mitomycin and tangcaopian piece of joint group of animals all survived, although not seen tang grass enhancement effect, but also no antagonism effect.(7) tangcaopian on a tumor-burdened animal blood cell count:the influence of the number of all kinds of blood cells in peripheral blood in mice solid tumor is lower than other groups, for unknown reasons. All kinds of blood cells in peripheral blood and ascites tumor mice number higher than that of mitomycin alone group, the difference only the red blood cells, the other groups (p<0.1)Conclusion:(1) Leukopenia mice model is established successfully with mitomycin.(2) Tangcaopian has the ability to increase white blood cells treated with mitomycin in vivo.(3) tangcaopian does not reduce the antitumor effect of mitomycin.
