Objective:Through detecting of the expression of NRAGE, HDAC3and NF-κB in esophageal squamous cell carcinoma tissues, analysis the relationship between NRAGE, HDAC3, NF-κB expression and the clinicopathologic characteristic of esophageal carcinoma, and the relationship between the expression of each of them and the radiotherapy curative effect in esophageal carcinoma tissues, preliminary explore the relationship between NRAGE, HDAC3, NF-κB and radioresistance in esophageal squamous cell carcinoma patients, and provide theoretical foundation for formulating reasonable and effective individualized treatment plan of esophageal carcinoma.Methods:Collecting27cases of esophagus squamous cell carcinoma tissue sample which were confirmed by fiberoptic esophageal speculum biopsy before radiotherapy. Protein expression level of NRAGE, HDAC3and NF-κB in tumor tissues were detected by streptavidin-peroxidase (SP) immunohistochemical staining method. All27patients with esophageal carcinoma were received conventional fractionated radiotherapy by6MV-X ray linear accelerator. Evaluated the therapeutic curative effect when radiotherapy completed and3months after radiotherapy referencing the esophageal barium meal evaluation standard. Using nonparametric test, spearman rank correlation and logistic regression method to analyze the results.Result:1The relationship between protein expression levels of NRAGE, HDAC3and NF-κB in esophageal tissues and the near future curative effect after radiotherapy In27cases of esophageal carcinoma tissues, the protein expression of NRAGE, HDAC3and NF-κB had different levels. Mean density value of NRAGE in the effective group was significantly lower than the invalid group, the difference was statistically significant (P=0.009). Mean density value of HDAC3and NF-κB in the effective group was lower than the invalid group, but it was not statistically significant (P=0.134and0.349).2The correlation among the protein expression of NRAGE, HDAC3and NF-κB in esophageal carcinoma tissuesIn27cases of esophageal carcinoma tissue, there was a positive correlation between NRAGE and HDAC3expression, the correlation coefficient was0.384(P=0.048). NRAGE and NF-κB, HDAC3and NF-κB protein expression had no obvious correlation(P=0.051and0.815).3The relationship between NRAGE, HDAC3, NF-κB protein expression levels and the clinicopathologic characteristic of esophageal carcinomaIn27cases of esophageal carcinoma patient, NRAGE, HDAC3and NF-κB protein expression level had no relationship with patient’s sex, age, lymph node metastasis status, lesion length and clinical stage, there were no significant statistically correlation (P>0.05).Conclusion:1The positive expression of NRAGE protein in esophageal carcinoma tissues was negative correlation with the near future curative effect after radiotherapy. Radiation effect in patients with higher NRAGE expression of NRAGE were better than those with lower NRAGE expression. It prompted that NRAGE may be a important indicator reflecting esophageal carcinoma radioresistance and the near future curative effect after radiotherapy.2In esophageal carcinoma tissue, there was a positive correlation between NRAGE and HDAC3expression. That is to say, there may be some relationship between NRAGE and HDAC3, this relationship may be influence radioresistance of esophageal carcinoma.3The expression level of NRAGE, HDAC3and NF-κB in esophageal carcinoma tissue had no relationship with patient’s sex, age, lymph node metastasis status, lesion length and clinical stage, there were no significant statistically correlation.
