OBJECTIVEFruit of Chaenomeles is commonly used as Chinese traditional medicine. It is reported to have various biological activities, including anti-inflammatory, analgesic, hepatoprotective, antibacterial, antitumor, antioxidant. Fruit of Chaenomeles is the dry and mature fruits of the Rose family Plant Chaenomeles speciosa(Sweet) Nakai, recorded in the China Pharmacopoeia(2010edition).Recent years, studies have found that not only the fruit of Chaenomeles can be used as medicine, its flowers, leaves, seeds also have some pharmacological activities. But studies on the seeds of Chaenomeles planted in Shandong Province have not been reported. Linyi areas in Shandong is one of the largest planting areas of Chaenomeles. But the seeds of Chaenomeles have no other purpose except for breeding, resulting in a waste of resources and pollution of the environment.In order to make better use of the Chaenomeles, we used chemomics as a guide to investigate the antioxidant activity of different polarity portions of the seeds of Chaenomeles and the qualitative and quantitative analysis of some chemical components.METHOD AND RESULTS(1)The Chaenomeles seeds planted in Shandong as research objects were used to study the best oil extraction method and oil extraction rate based on orthogonal experimental design of three factors and three levels. The results showed that the oil extraction rate of the Chaenomeles seeds (about20%) is similar to those of Papaya seeds, lemon seeds, pomegranate seeds and apple seeds.(2) GC-MS technology was used to analysis the major fatty acid composition of the Chaenomeles seeds. The possible components of the11common chromatographic peaks were tentatively identified. The results showed that the content of unsaturated fatty acid was up to87%.Of all the fatty acid, the content of monounsaturated fatty was55.186%, polyunsaturated fatty was31.893%.And the components of the oil by Soxhlet extractor method and traditional press method were basically the same.(3) Using ultrasonic extraction method and the solvent extraction method, combined with HPLC chromatogram, the extract of the Chaenomeles seeds was divided into several different polarity parts. The antioxidant activity and total polyphenol content of different polarity parts were determined by the method of DPPH and ABTS. The results indicated that the ethanol extract showed the strongest antioxidant activity and the highest polyphenol content.(4) Using silica gel column chromatography. thin layer chromatography and HPLC-VWD-MS technology to identify several components from the effective portion. The possible4components characterized by chromatographic peaks were tentatively identified as myristic acid, hexadecanoic acid, pentadecanoic acid, dodecanoic acid. Another compound was similar to antioxidant2246, but it was not accurately determined.CONCLUSIONThis subject selected seeds of Chaenomeles Planted in Shandong which has not yet been researched as the research object, and adopted the Chemomics as the guiding idea to make an overall research of the Chaenomeles seeds, which has an important meaning on promoting the comprehensive utilization and modern research of the seeds of Chaenomeles Planted in Shandong.
