Vitis amurensis,belongs to Vitis Plant of vitaceae,The plant is widely distributed in china and rich in resource. Vitis amurensis main used saccharomyces,in the course of making wine,it can produce large amount of grape seeds, and its main components are polyphenols.At present,there are many researhces on the polyphenols internationly,and there has been a remarkable rate of Progress, but the study of vitis amurensis polyphenols far less belong to plant grape polyphenols and other plants .Research shows that polyphenols content in Vitis amurensis seeds far more than Vitis amurensis peels ,flesh and other planting grape varieties.Polyphenols are widely applied to Pharmaceutical,food and cosmetic industries etc,since they contain a variety of biologieal activities,such as anti-oxidation,anti-freeradieals, anti-ultraviolet,anti- virus, anti-inflammatory and so on.If we can change waste into valuables and make further Proeessing of the Plants,there will be important economic value and praetical significance.In this paper,Vitis amurensis seeds after making wine was taken as material,the fllowing contents were mainly studied:1. The ultrasonic technology applied to the extraction of vitis amurensis seed polyphenols.The effects on yield of ethanol concentration,ultrasonic temperaturre, ratio of solid/liquid,ultrasonic time,and ultrasonic times were researched by single factor assays.Then the optimum condition of the extraction used ultrasonic technology was carried out through orthogonal test design of L9(34)and variance analysis, comprehensive consideration of the extraction and production cost, to optimize the extraction conditions,and with the traditional solvent extraction were compared.2. The Present studies implemented macroporous resin to purify polyphenol extracts of Vitis amurensis seeds,bearing the merits of simple instrument,stable quality,low cost and easy to operation. By static adsorption and desorption experimental investigated AB-8 macroporous resin on adsorption,desorption performance of Vitis amurensis seed polyphenols,and the static and dynamic adsorption and desorption conditions and influencing factors of concrete research.3. The method of polyphenols content assay in Vitis amurensis seeds extracts was obtained by the research of UV-vis spectrophotometry,color conditions, color temperature and time colour-display reagent,and inspecting the methodology. By this method,the content of total polyphenols was determined in different seed extracts.4. The antioxidation activities of polyphenol extracts of Vitis amurensis seeds as well as Vc were studied by different systerms,including the removal of superoxide anion experiments, the removal of hydroxyl radical experiments, DPPH free radical scavenging and reducing power test experiments.Through a great deal of experiments,we can darw conclusions as follows:1. Using the ultrasonic technology ,vitis amurensis seed polyphenols after making wine was extracted.Orthogonal test results indicated that extraction temperature have strong influencest to the extraction eeffets of the 4 factors in the experiments,it includs extraction time,material-.liquid ratio and extraction times.The influencing extents of the 4 factos are: ultrasonic temperature> material-liquid ratio> ultrasonic time> ultrasonic times. On the baisis of single factor assays and orthogonal test, The optimum condition of the extraction was carried out: ethanol concentartion is 60%,temperature is 50℃, extraetion time is 30minutes , ultrasonic power 120W, solidus-Liquid ratio is 1:8 and extraetion times is 2. Using ultrasonic wave extraction, the yield was higher than the traditional organic solvents extraetion,Considered time and resources using,ultrasonic wave extraction method had more advantages than organic solvents extraction method.2. Using AB-8 macroporous resin, crude extractions of vitis amurensis seeds were purified.The results showed that the best adsorption conditions and desorption conditions with AB-8 macroporous resin were as follows:the sample concentration is 10mg/ml,the moving speed is 3ml/min,eluting volume is 2BV , eluting solvent is 60% ethanol solution and the eluting speed is 2ml/min. In this contidation,the Puriy of polyphenols of extracts of Vitis amurensis seeds can be improved from 35.02% up to 88.8%. 3. Using Folin-Cioealteu colorimetry,the content of polpyhenols in Vitis amurensis seed extracts is detemrined. Through contrasting colors conditions,the best detemrination condition as ofllows:1ml sample solution, 3ml distilled water, FC colour-display reagent 1.5ml, 20% sodium carbonate solution 3ml, With the capacity to 10ml distilled water, shaking, room temperature to avoid light placed 2h, in 760nm determination spectrophotometry.Then,inspecting the content determination of methodology, the results:There was a good linearity between gallic acid concentration and absorbance in the range 0.002-0.01 g·L-1(r = 0.9996);the precision RSD was 0.14%;the Stability RSD was 1.77%;the repeatability RSD was 1.44%;The average recovery rate was shown to be 102.00%(RSD=1.37%). Conclusion :This method is simple,fast and has high sensitivity and goodreproducibility.4. Antioxidation activities results showed that their IC50 were differ from systems, crude extraction and purification of polyphenol of Vitis amurensis seeds both had the antioxidating function. crude extractings of Vitis amurensis seed polyphenols after purification, the ability of free radical scavenging and reducing power were increased significantly.polyphenols purification of Vitis amurensis seeds reductive ability and the ability to clear a DPPH free radical stronger than Vc, and the ability in the clear hydroxyl radicals and superoxide anion weaker than Vc. Polyphenol extracts of vitis amurensis seeds with the antioxidant activity can make the good natural antioxidants,with the research and development of high value.The experiment has comprehensive studied on polyphenol extracts from Vitis amurensis seeds. Not only for the exploitation and utilization of vitis amurensis seeds after brewing provides basic research reference and scientific basis and helps all aspects of the in-depth study in vitis amurensis seeds, to further research, it also promotes the sustainable development of vitis amurensis industry.
