Objective To study the expression of E-cadherin, TGF-β1and Twist protein and investigate its significance in the occurrence and development of prostate cancer.Methods The expression of E-cadherin, TGF-β1and Twist protein in59prostate cancer tissues and21benign prostatic hyperplasia (BPH) tissues were detected by immunohistochemical SP staining, and the correlation with clinico-pathological features and the correlation between three were analyzed.Results1. Positive rates of E-cadherin, TGF-β1and Twist were32.2%,71.2%and74.6%, respectively, in prostate cancer tissues and90.5%,9.52%and0, respectively, in BPH tissues, with significant differences between the two groups (P<0.05).2.The reduced expression of E-cadherin was related to the differentiation of prostate cancer tissues and PSA level, but not associated with clinical stage, lymph node metastasis, bony metastasis and age. The increased expression of TGF-β1and Twist was related to the differentiation of prostate cancer tissues, clinical stage, lymph node metastasis, bony metastasis, but not to age. The positive expression of TGF-β1was significant differences between PSA<20μg/L group and PSA>20μg/L group, but the positive expression of Twist was not a significant between-group difference.3.The expression of E-cadherin was highly negatively correlated with that of Twist. The expression of TGF-β1was correlated with those of E-cadherin and Twist.Conclusion1.Compared with BPH, low expression of E-cadherin in prostate cancer and its intimate correlation with the differentiation of prostate cancer tissues and PSA level indicated that it participated in the occurrence and progression of prostate cancer. But it was not as the independent factor to judge prostate cancer’s progression and prognosis.2.Compared with BPH, high expression of TGF-β1and Twist in prostate cancer and its intimate correlation with the differentiation of prostate cancer tissues, clinical stage and metastasis indicated that it may play important contribution in the occurrence and progression of prostate cancer. It could promote the invasion and metastasis of prostate cancer.3.The hallmark event of the occurrence of epithelial-mesenchymal transition was the reduced expression of E-cadherin. TGF-β1and Twist have synergistic effect in regulating the course of epithelial-mesenchymal transition. Epithelial-mesenchymal transition was an important phenomenon in the occurrence and progression of prostate cancer. It plays an important role in prostate cancer because of increasing the migrated ability of cancer cell. And epithelial-mesenchymal transition could a reasonable explanation of the mechanism of the initial steps of the invasion and metastasis of prostate cancer.
