Objective:By detecting the protein expressions of Twist, E-cadherin, N-cadherin in supraglottic laryngeal carcinoma to investigate the their roles in the occurrence, progression, and prognosis of supraglottic laryngeal carcinoma.Method:Use immunohistochemical method to detect the expressions of Twist, E-cadherin, N-cadherin protein in46cases of supraglottic laryngeal carcinoma and30cases of polyp of vocal cord.Results:1.In the supraglottic laryngeal carcinoma tissues, the positive expression rates of Twist, E-cadherin and N-cadhrein protein were69.57%,39.13%,50.00%respectively, while in the polyp of vocal cord tissues, they were36.67%,86.67%,13.33%respectively. The differences of the three indicators between laryngeal cancer tissues and benign laryngeal lesions tissues had statistical significances (p<0.05).2.The positive expression rates of Twist and N-cadherin in the supraglottic laryngeal carcinoma tissues with low grade, with lymph node metastasis, T3+4, late stage were higher than those of high grade, without lymph node metastasis, T1+2, early stage (p<0.05). While the positive expression rate of E-cadhein in the the supraglottic laryngeal carcinoma tissues with low grade, with lymph node metastasis, T3+4, late stage was lower than that of high grade, without lymph node metastasis, T1+2, early stage (p<0.05).3.The expression of Twist negatively correlated with the expression of E-cadherin in the supraglottic laryngeal carcinoma tissues (r=-0.535, p=0.000); The expression of Twist positively correlated with the expression of N-cadherin in the supraglottic laryngeal carcinoma tissues (r=0.378, p=0.010); The expression of E-cadherin negatively correlated with the expression of N-cadherin in the supraglottic laryngeal carcinoma tissues (r=-0.445,p=0.002);4.The local recurrence and neck recurrence were higher in patients with Twist, N-cadherin positive expression than those in patients with Twist, E-cadherin negative expression (p<0.05) in3years. While the local recurrence and neck recurrence were lower in patients with E-cadherin positive expression than those in patients with Twist, E-cadherin negative expression (p<0.05) in3years.5.The3-year overall survival rate and3-year disease free survival rate were lower in patients with Twist, N-cadherin positive expression than those in patients with Twist, E-cadherin negative expression (p<0.05). While the3-year overall survival rate and3-year disease free survival rate were higher in patients with Twist, N-cadherin positive expression than those in patients with Twist, E-cadherin negative expression (p<0.05).6.The results of multivariate Cox proportional hazards model confirmed that only the presence of cervical lymph node metastasis and the depth of tumor invasion are the significant, independent predictors of prognosis (HR=8.639,4.276,p=0.037,0.023).Conclusion:1.Twist, E-cadherin, N-cadherin protein may be correlated with the occrrence of the supraglottic laryngeal carcinoma.2.Twist, E-cadherin, N-cadherin protein may be correlated with the invasion and metastasis of the supraglottic laryngeal carcinoma.3.Twist protein presumably correlates with epithelial-mesenchymal transition in supraglottic laryngeal carcinoma.4.Twist, E-cadherin, N-cadherin protein may be correlated with the prognosis of the supraglottic laryngeal carcinoma.
