Part one Comparison of two kinds of preparation of rat HCC modeltrasplantation methodObjective:Comparion of cell suspension method and mass embeddingmethod in preparation of rat models, select the appropriate method of rat livercancer modleMethods:160Wistar rats, male,6weeks old145g-160g weight. CBRH-7919tumor. We choose open transcatheter injection of cell suspension method andabdominal tumor mass embedding method to establish the animal of25and35respectively. Calculated operation time, the rate of tumor formation.2the rats were under MR T1and T2sequence scanning after7days,10days,14days after operate respectively, to observe two kinds of modelingmethod in tumor size, shape, distance, necrosis of the tumor. Three rats werekilled randomly, taken the tumor mass and liver tissue, observed tumorgengeral,fixed and paraffin blacks embedding,then conventional slice, HEstaining, CD34and VEGF were detected by immunohistochemistry.Rusult:1The cell suspension method planted a total of25rats, a total of43tumors, operation time was25.2±4.1minutes, average blood loss is less than0.2ml,14days after the operation there were five rats dead, three rats notumor block, two rats had distant metastasis, modeling success rate of60%(15/25), tumor formation rate of72.09%(31/43). Tumor mass planting of35rats, a total of62tumors, operation time was10.13±2.01minutes, averageblood loss of about0.4ml,14days after the operation there were3rats dead,two rats with no tumor mass formation, ratsdistant metastases, modeling thesuccess rate of88.57%(31/35), the tumor was88.07%(54/62). 2Tumor specimen section view as an fish-like off-white, unclearboundaries and the liver. HE staining showed that the CBRH-7919tumor cellswere nest-like distribution, cells arranged in disorder, abnormal high,triangular or irregular in shape, the nucleus is large and the color depth. VEGFstaining of tumor neovascularization colored visible within the nests of brownspot.CD34staining slice scattered deeply brown in the tumor area, VEGFtumor area cell pack slurry deeply stained.3MR scan7days after the suspension method group there were no cleartumor mass at all, developing10days there were three rat liver showedmixed-signal patchy, ill-defined,14days after17rat liver visible shape isnotrules, tumor mass total31,0.74±0.52cm,4large tumor masses wereseen in the liquefied necrotic area. The tumor block group7days after MRscan round more uniform size of the tumor masses,0.53±0.32cm, internal noliquefaction necrosis.Conclusion: Tumor mass embedding method have higher rate of tumorfomation, lower necrosis rate,what’s more it can into the tumor in the earlydays. And the correlation betweenthe various perfusion parameters and immunohistochemistry would beanalyzed.Materials: To meet the requierments from the preparation of a rat modleof screening. A total of34tumor-bearing rats tumor size average0.57±0.34cmwere divide into two group, the experimental group were26,the control groupof8.Siemens Experimental group PO solafinb20mg/d, control group PO NS 20mg/d,for21days。Dual-source CT perfusion scan mode perfusion scan intwo groups of rats before and after treatment, diluted alcohol(300)1ml(iohexol: saline=1:5) injected through the femoral vein at rate0.5ml/s into therat, delay2second to scan, records all of the perfusion parameters. The end ofthe second scan eagerly rats were sacrificed, tumor tissues were fixed in10%neutral formalin for36-48hours, embedded in paraffin blocks, slicesunderwent HE and immunohistochemical examination.Result:1normal liver tissue time-density curve slow rise and drop type, take theform of natural curve;HCC Time-density curve(TDC) performance: rapidlyrise in the arterial phase, decline rapidly after peak。2CT perfusion parameters of the experimental group tumor areastatistical results are shown: the area of liver cancer in experimental ratsbefore and after treatment paired T-test found BF, BV, pBV, ALP weresignificantly different (P <0.05), while the MPI, PVP, HPI value were notstatistically different (P≥0.05). Before and after treatment in the controlgroup tumor area MIP, BF, BV, PBV, ALP, PVP and HPI equivalent PairedT-test P values were greater than0.05, not statistically significant.3Changes in maximum diameter of the experimental group is-0.302±0.234cm,and the control group is0.692±0.536cm, Changes in value of themaximum tumor diameter of two groups of rats before and after treatment wasstatistically significant, P=0.014(P <0.05).4After solafinb treatment, the number of MVD is decreased than treat byNS, the two is a statistically significant (P=0.021, P<0.05);NS group VEGFis expression and compared with solafinb group was statistically significant(P=0.015, P<0.05). MIP with VEGF and MVD was a significant positivecorrelation respectively (r=0.521and0.517, P <0.05); ALP with VEGF andMVD was significantly positive related(R value of0.647and0.568,respectively). Conclusion: Dual-source CT can analysis the hemodynamic changes ofHCC after treatment with solafinib quantitatively...
