Tree shrew is a newly important experimental animal. It has more closed phenogenetic relationship with primate than other small animals. Because of its smaller body size and shorter reproductive cycle, it has been a great animal of disease models used in the research of human diseases. However, the absent of its genotype background has been an major obstacle in the broad application of tree shrew as animal models.High mobility group box1(HMGB1), Interleukin4(IL-4), Interferon-gamma (IFN-γ), and Caudal homeobox gene (CDX2), which have very important function in vivo in other species, have not been reported in the tree shrew. In this thesis, we cloned the gene of HMGB1, IL-4, IFN-γ, and CDX2of the tree shrew. That would lay a solid foundation in the study of their function in the tress shrew in the future.In the thesis, mainly through the tool of molecular biology, we got the following results:1) we cloned the full coding sequence of the gene HMGB1, which has648bp, gene IL-4, which has426bp, and gene IFN-γ, which has501bp. We got709bp of nucleotide sequence of the gene CDX2.2) Through sequence similarity alignment, we found there is99.5%similarity between tree shrew and primate in the gene of HMGB1,72.9%in the gene of IL-4, and76%in the gene of IFN-γ. In phylogenesis analysis, we found tree shrew is more closed to primate than other species.3) we expressed the recombinant protein of HMGB1and got purified form which is ready to study the biological function.Overall, we fist cloned the full coding sequence of the gene of HMGB1, IL-4, and IFN-γ of tree shrew, which is important in the study of those gene function in tree shrew. We got the recombinant protein of HMGB1, which could be used to study the biological function in tree shrew. We also got part of the gene coding sequence of gene CDX2, which might be a clue in the further clone and its functional research.
