In the current industrial production, cowhide offcuts, which were considered as waste, were massively produced and thrown away, resulting in a serious waste of resources and environmental pollution. In this paper, the cowhide offcuts were used as raw materials and hydrolyzed by pancreatic enzyme. The hydrolysing process was optimized through response surface analysis. Then, the influence of external conditions on the viscosity of the collagen hydrolyzate and the antioxident capacity of the cowhide collagen were investigated. The approach of cowhide offcuts hydrolysis and extracting collagen from them was built up initially. And certain data support for further research&development of these resources was provided.From the beginning, the cowhide offcuts were pretreated for decontamination and then hydrolyzed by pancreatic enzyme. After comparing with ninhydrin color reaction, formol titration method was chosen to characterize the degree of cowhide collagen hydrolysis. Impact factors were determined through factor analysis and then Expert-Design7.0software was employed to conduct the response surface analysis. The process conditions of pancreatic enzyme hydrolysis of cowhide collagen were optimized.And then the influence of the solution concentration, the external temperature, pH value and the storage time of the sample on the viscosity of cowhide hydrolyzed collagen solution are analysed, The analysis results of the response surface method indicated that:when the concentration of sample was0.75mg/mL, the temperature is40.30℃, pH value is7.50and the storage time is48.00h,the viscosity of cowhide collagen solution was to achieve maximum in theory.Finally the determination of reduction force combined with DPPH free radical scavenging method were used to investigate the antioxidant ability in vitro of cowhide collagen. The results showed that the clearance rate of cowhide collagen to DPPH free radical increased as the increased amount of protein, the clearance rate reached74.76%when protein/DPPH methanol solution was5mg/ml, and the half inhibiting concentration EC50was1.63mg/ml; The determination results of reduction force also showed that, the higher the concentration of sample protein, the stronger its reduction force.
