A central composite design(CCD) was applied to build up a polysaccharidesextraction model and to search an optimum extraction model for OPHiocordycepssinensis (CS) mycelium. Four factors are chosen, containing solvent PH (3-11),extraction temperature(40°C-120°C), extraction time(40min-180min) andvolumes(10-30). All these factors were divided into five levels. Experiment resultsshow that only extraction temperature has a significant influence on the yield. Allthese process carried out by the software of design-expert7.0.There are a lot of starch exist in the cultured of mycelium of O. sinensis, sodesign to remove the starch from mycelium should be done before the research of O.sinensis polysaccharides. Apply α-amylase to this project, measure the content ofreducing sugar by DNS. To prove the content changed not significance, actuallyfrom6h to9h the content of reducing sugar is same mainly and it proved starch isremoved.30trails were employed to build this model, calculated by the software, aequation is given and work out the optimal scheme. The optimum conditions are PH3, extraction time110min, volumes10, and extraction temperature120℃,and atthis optimum conditions69.5854mg polysaccharides can be obtained. To test it infact, predicted values were same to real values.In the other hand,model wouldevaluate the result through R-Squared, Adj R-Squared, Adeq Precision,PredictedR-squared.All these evaluation can ensure the model is well.Compare all these factors and get the result that only temperature has thesignificance of the result of polysaccharides quality in the model.The research of immune actives were provided by mouse spleen cellproliferation from splenocyte in vitro. The result suggests that polysaccharidesextraction have effect on immune actives in lymPHocyte proliferationassay,especially the concentration at100μg/ml with2μg/ml ConA.To compare the character of different condition, we study the PHysicochemicalproperty collected in two different conditions.The study contains four aspects: Monosaccharide Formation, Polysaccharide molecular weight, IR and the rate ofpolysaccharides in extraction. In the part of Monosaccharide Formation, mainly theyboth contain Man, Glc and Gal. In the part of Polysaccharide molecular weight andIR,they are nearly same. In the part of rate, one result is13%and the other one is37%.
