| Dictyophora indusiata is classified to Dictyophora of phallaceae,an edible fungus and precious drug,which widely distributed in Fujian,Jiangxi,Yunnan and other parts of China.Modern pharmacological studies have shown that Dictyophora polysaccharides has anti-inflammatory,antihypertensive and antineoplastic.In order to further expand its applicable function,this study was focused on the extraction technology of Dictyophora polysaccharides and its antioxidative properties,the proliferation effects of Dictyophora polysaccharides on Bifidobacterium adolescentis in vitro.The results are as follows:1.Based on the single factor experiment,dictyophora is used as raw material and the optimum extraction experiment of polysaccharides by the combination of ultrasonic and microwave was carried out.The results have showed that under the following conditions:microwave power of 150W,ultrasonic power 550W,the ratio of material to liquid 45mL/g,extraction time 6min,the yield of Dictyophora polysaccharides could reach 12.36%.2.The four extraction methods(UMAE,HWE,UAE and MAE)were comparatively analyzes in experiment influences on antioxidant of Dictyophora polysaccharides.The results are as follows:Antioxidant of polysaccharides extracted by different extraction methods were showed.The polysaccharides extracted by UMAE had higher DPPH-,OH·and superoxide anion radical scavenging capacity.3.The antioxidant activity and the prebiotic activity of different fractionated alcohol-precipitated fragments were studied by ethanol fractionation on extraction of Dictyophora polysaccharides.The results have showed that DPPH-,OH·and superoxide anion were showed by using ethanol water mixture with different volume fraction(20%,40%,50%,60%and 80%,respectively).OH-,the superoxide anion radicals are stronger,with increasing of the ethanol concentration while the antioxidant capacity decreased in the group.The infrared spectroscopy showed that the ethanol precipitation process haven’t changed the structure of Dictyophora polysaccharides;The proliferation and anti-hydrolysis ability of polysaccharides to B.adolescentis increased with the increase of ethanol concentration,which indicated that Dictyophora polysaccharides has better proliferation effect on Bifidobacterium adolescentis and has the potential of prebiotics.4.The optimal recipe of the polysaccharide tablet was optimized on the basis of the single factor test and the sensory evaluation score as the index.The optimum formulation are as follows:0.12g of citric acid,0.08g of xylitol,0.15g of starch and 0.03g of microcrystalline cellulose.The product quality standard of Dictyophora polysaccharides slice was established in this dissertation. |
