Spiranthes lancea is the root or the whole grass of orchid plant Spiranthes grass.Main research target is extraction and purification of Spiranthes polyphenols.Establish-ed optimum methods of Spiranthes lancea polyphenols analysis.Established optimumprocess for Spiranthes lancea polyphenols extraction with microwave assisted.Analyzedthe composition of Spiranthes lancea volatile oil.Main conclusions are follow:Established optimum colorimetric conditions of spiranthes lancea polyphenolanalysis:Folin-Ciocalteu reagent was l.5mL,volume ratio between Folin-Cioealteureagent and10%Na2CO3was1:2.5,reaction time was2h,temperatue was35℃,detection wave was760nm,average recovery rate was103.12%.It is accurate andreliable for spiranthes lancea polyphenols detection.Established optimum process of Spiranthes lancea polyphenol extraction withmicrowave assisted:extraction process were optimized on single factor experiment byapplication of response surface analysis method.Optimum process of polyphenolextraction is: ethanol concentration of47%,time of84minutes, extraction temperature of48℃,Spiranthes lancea polyphenol producttivity reached5.41mg/g.Established macroporous resin purification process of Spiranthes lanceapolyphenol.D101macroporous resin was established to be the most suitable forSpiranthes lancea polyphenols purification after comparision five kinds macroporousstatic and dynamic adsorption.Dymatic adsorption condition was: polyphenol pH with5.10,adsorption solution concentration with1.60mg/mL,flow velocity was1.50mL/min,maximum dymatic theory adsorption amount was35.04mg/g.Polyphenol purity reached23.26%after desorption.Analyze Spiranthes lancea volatile oil component using GC-MS method at firsttime.From Spiranthes lancea oil composition comprehensive analysis and study,27components and their relative amounts were established,identified componentsaccounted for95.32%of the total peak area.
