Recent years phlorotannin as a marin plant polyphenol attracted widespread attention,China had abundant resources of phaeophyta.This paper taked sargassum fusiforme of kelp as material.This paper study the sargassum fusiforme polyphenols ethanol extraction,macro resin column chromatography process,extraction main components were separated by semi-preparative HPLC,using HPLC-MS preliminary analysis of its structure,and study free radical scavenging in vitro,anti-H2O2 hepatocytes oxidative damage and antioxidant activity in mice to macro resin purification compound.The optimal conditions was optimized RSM as the factor experiments.Extraction temperature 40℃,alcohol concentration 25%,times 3 h,ratio of liquid to solid 30:1,under this condition the polyphenols content was 8.84 mg/g.It was no significant difference to the theoretical value only 1.37%,the model was feasible.By Column chromatography experiments showed that resin D-101was preferable for separation and purification of sargassum fusiforme polyphenols.The flow of sample rate 1 BV/h,the concentration of sample was 2.4 mg/ml,using 50%ethanol as eluant about with 5 BV,the flow rate of desorption 3 BV/h,this were the best purification condition.Under this condition purification and separation of sargassum fusiforme polyphenols purify of about 32.75%.There main components were purified by semi-preparative HPLC,C10H3O15、C6H4O2、C8H3O8 were formula of three components by septrum sepeculate.Sargassum fusiforme polyphenols had a good scavenging for DPPH and·OH.The IC50values were 4.57 mg/L、13.63 mg/L.The total antioxidant capacity was 65.56 U/mg.This showed that sargassum fusiforme polyphenols had very strong antioxidant capacity.The cell experiments results show that Sargassum fusiforme polyphenols can significantly increase within RBL SOD(p<0.05),CAT(p<0.01),GSH-PX(p<0.01)activity,decreased MDA content,reduced damage H2O2 cells LDH release.Results showed obvious increase in protective effection on H2O2 damaged liver cell.Through animal experiments showed that sargassun fusifome polyphenols can significantly increase the antioxidant capacity in mice,Compared with the model group Sargassum fusiforme polyphenols can significantly reduce MDA in mice,(p<0.01),all experiments group can reduce serum MDA content(p<0.01),only high dose groups was significantly in liver(p<0.01);all dose Sargassum fusiforme polyphenols can be significantly improved serum GSH-PX activity in three groups(p<0.01),high dose group could significantly increase the liver GSH-PX activity(p<0.01);middle and high dose groups can significantly increase serum and liver SOD activity(p<0.01);middle and high dose Sargassum fusiforme polyphenols have a good inhibition of protein carbonyl content(p<0.01).
