Celastrus angulatus Maxim is a perennial plant of insecticidal activity, which is belonged to Celastraceae, Celastrus L. This plant is distributed over the basin of the Yellow River and the Yangtze River widely, and it is a very valuable resource to research and develop botanical insecticide. There are many kinds of insecticidal compositions in the root bark, the seeds and the leaves, most of them are sesquiterpene polyol esters bearing β-dihydroagarofuran skeleton. Compounds of this species have several kinds of activity to insect, such as poisonous activity, antifeedant activity, anesthesia activity and so on. Because most former research on Celastrus angulatus focuses on the root bark, in order to make further research, improve the utilization efficiency of natural resource and realize sustainable development, we have undertaken a systematic investigation on the seeds of Celastrus angulatus, especially, its chemical constituents.In this paper, extract of the seeds of Celastrus angulatus, seeds oil, was obtained by circumfluence distillation, with petroleum ether as solvent. After being extracted by methanol, the extract was divided into two parts. Then with petroleum ether-ethyl acetate as eluents, both of the concentrate of two parts were isolated through silica gel chromatography column at atmospheric pressure, respectively. After being isolated and purified ulteriorly by preparative high-performance liquid chromatography, eight compounds of high purity were obtained. Using a combination of several kinds of1D and2D NMR techniques, which includ in1H NMR,13C NMR, DEPT,1H-1H COSY, HSQC and HMBC, the structures of those compounds were identified. They are Angulateoid B (Y-1),1β,2β,8a,15-tetraacetoxy-9a-benzoyloxy-β-dihydroagrofuran (Y-2),1β,8β-diacetoxy-9β-benzoyloxy-β-dihydroagrofuran (Z-1), Angulatueoid C (Z-2), Angulatueoid H (Z-3),1β,15-diacetoxy-8β-hydroxy-9β-benzoyloxy-β-dihydroagrofuran (Z-4),1β,2β,8β,15-tetraacetoxy-9β-benzoyloxy-β-dihydroagrofuran (Z-5), Angulatusine A (Z-6). Four of them, Y-2, Z-1, Z-4and Z-5are new compounds. The1H NMR and C NMR spectra of Angulatueoid B, Angulatueoid C, Angulatueoid H and Angulatusine A were completely assigned, too. Angulateoid C is one of the most important compounds of the seeds of Celastrus angulatus, and the insecticidal activity of this compound is very high. So far, there is no report on separating and purifying this compound from the seeds oil of Celastrus angulatus by macroporous resin. In this paper. Angulateoid C was separated and purified from the seeds oil of Celastrus Angulatus by macroporous resin for the first time. Four types of macroporous adsorption resin were studied to compare their ability of adsorption and desorption for Angulateoid C, and the result indicated that HP20resin was the best. The influence of sample quantity, the concentration and volume of elution on the purification ability of HP20resin was observed and compared, and the appropriate conditions were optimized. The optimum process was that sample of3times macroporous resin in quantity was absorbed by column of HP20macroporous resin, the column was washed with40column volume of40%ethanol first, and then90column volume of60%ethanol. After being purified, the content of Angulateoid C is raised more than7times. This method offers a very important scientific foundation for producing preparation of the seeds of Celastrus angulatus in industry.
